Propofol is an anesthetic which is used to initiate and maintain general anesthesia or sedation. It is widely used for surgical procedures due to its short acting time and its reversibility. Recent reports indicate that an increasing number of people are addicted to propofol owing to the feeling of “a good sleep” after propofol-induced anesthesia. After surgical procedures, such as conscious sedation endoscope, many patients experience memory impairment and delirium accompanied by a serious cognitive function deficit. Thus, it is necessary to investigate the medical and psychiatric effects of propofol on sleep architecture. The stages of sleep are wake, non-rapid eye movement (NREM) sleep, and rapid eye movement (REM) sleep. These stages are differentiated by distinctive electroencephalogram (EEG) patterns. Specifically, NREM sleep is characterized by high levels of delta waves, with sleep spindle waveforms and slow wave formations. REM sleep is primarily characterized by low amplitude brain rhythms in theta wave formations. I investigated sleep patterns and brain rhythms in propofol-injected mice and found changes in both NREM and REM sleep stages occurring for 1 hr post propofol injection. Following propofol injection, the total duration of NREM was substantially increased, particularly in SWS1 (light sleep), and the total duration of REM was significantly decreased. In addition, I implemented a novel object recognition test to investigate the degree of memory impairment induced by propofol. The propofol treated mice did not discriminate objects well compare to saline injected mice. Memory impairment following propofol injection was correlated with the frequency of sleep spindles in SWS1 and decreased REM sleep. However, memory tests performed 24 hours after propofol injection with Y-maze confirmed that no memory damage was caused by propofol. These results indicate that effect of propofol is short. In addition, I experimented an effect of long-term injection of propofol. The total duration of long-term propofol dose group in light- and dark-phase were significantly changed. The maintenance of SWS1 was increased and incidence of SWS2 was decreased. Future research should investigate the long-term effect of propofol on memory. And the short-, and long-term effect of propofol on physiological and molecular experiments in mice.
프로포폴은 마취 상태를 만들거나 지속적으로 유지시키거나 혹은 진정제로 널리 쓰이는 마취제이다. 프로포폴은 지속되는 그 효과가 짧기 때문에 이런 이유로 간단한 시술이나 수술에 널리 사용되고 있다. 최근에 이러한 프로포폴에 중독증상을 보이는 환자들이 늘어가고 있는데, 그것은 바로 프로포폴로 인해 안정된 휴식 혹은 숙면을 취했다고 생각하는 사람들이 늘어가고 있기 때문이다. 그러나 보고에 따르면, 프로포폴이 기억에 손상을 일으킨다는 연구가 보고되고 있다. 이러한 문제들로 인해 프로포폴이 수면패턴에 끼치는 영향에 대한 연구가 필요로 하고 있다. 수면은 크게 각성상태, NREM 수면, REM 수면으로 나뉘어 질 수 있으며, 수면의 각 단계들은 각각 다른 뇌파 (EEG)의 양상을 띠고 있으므로 뇌파분석을 통해서 나눌 수 있다. 특히, NREM 수면의 경우 델타 파장 영역의 뇌파가 강하게 나타나는데, 이 느린 델타파에서 sleep spindle이 나타나는 것이 특징이다. 반면 REM 수면은 빠르고, 작은 진폭의 리듬인 세타파가 많이 나타나는 것이 특징이다. 사람의 수면패턴과 생쥐의 수면패턴은 유사하기 때문에, 프로포폴에 의해 변화하는 수면의 양상을 생쥐에서 특히 NREM 수면과 REM 수면에 집중하여 연구할 수 있다. 프로포폴을 투여한 후 생쥐의 수면을 리코딩 하였으며, 분석결과 얕은 잠을 많이 자는 결과가 있었으며, 그리고 이러한 약물에 의해 변화한 수면이 기억에는 어떤 영향을 끼치는지에 대해서도 자세히 연구하기 위해 Novel object recognition task를 진행하여 생쥐의 기억에 어떤 변화가 있는지도 연구하였다. 연구결과 기억이 손상되었음을 확인할 수 있었다. 이 후 연구로는 프로포폴을 장기적으로 주사하였을 때 변화하는 수면의 양상과 그에 따른 기억에 어떤 영향을 미치는지 연구할 필요성이 있다.
