최근 미세소관 세포골격을 표적으로 삼아 항암제 개발에 대한 관심이 높아지고 있다. 본 연구에서 우리는 세포 내 및 세포 외 미세소관 네트워크를 표적으로 삼고, 특히 β-튜불린 표면의 결합 부위를 표적으로 삼는 일련의 벤즈이미다졸 유도체를 개발했다. 화합물 BE7-7 및 O7-7은 테스트된 다양한 세포주에서 인상적인 항증식 활성을 나타냈으며 각각 15.74 및 33.15의 선택 지수를 나타냈다. 암 세포주의 경우 BE7-7은 1.42 ~ 7.8μM의 IC50 값을 달성한 반면, O7-7은 1.13 ~ 8μM의 IC50 값을 나타냈다. BE7-7 및 O7-7은 상처 봉합에 대한 상당한 억제를 나타냈으며, 상처 봉합 비율은 0μM에서 37-38%에서 0.43%로 감소하고 20 μM에서는 0% 미만으로 감소했다. 놀랍게도 콜로니 형성 감소율은 BE7-7이 95.8%, O7-7이 99.97%로 현저히 감소했다. 이러한 결과는 BE7-7과 O7-7이 암세포 이동을 방해하는 능력이 있고 복제를 억제할 가능성이 높다는 것을 나타낸다. 세포주기 중단은 G2/M 단계에서 세포주기 중단으로 이어졌으며, 이는 BE7-7과 O7-7이 미세소관의 불안정화를 통해 항암 활성을 나타냄을 나타낸다. 또한, 세포 내 이미징을 통해 암세포 내 튜불린 네트워크 억제 효과를 관찰했다. 전반적으로, 우리 연구는 합성된 벤즈이미다졸 유사체의 신규하고 선택적 항암제로서의 잠재력을 입증하였다. 우리는 또한 천연 화합물인 유제놀의 Schiff 염기 유도체이며 세포 내 양이온을 검출하는 두 개의 이미징 프로브인 FHE와 FLHE를 개발했다. 이러한 파생물은 모두 생물학적 및 환경적 샘플에서 Zn2+ 검출에 대해 높은 특이성을 나타냈다. FHE와 FLHE는 분자내 전하 이동(ICT)으로 인해 낮은 형광 신호를 나타냈다. 그러나 HEPES 완충액(pH = 7.4, 50% ACN, v/v)에서 Zn2+ 이온을 사용한 적정은 매우 특이적인FHE-Zn2+ (Ka = 1.1 × 104 M-1) 및 FLHE-Zn2+ (Ka = 5.33 × 103 M-1) 복합체가 형성된다. 이로 인해 CHEF(킬레이트화 강화 형광) 효과로 인해 ICT가 중단되고 그에 따른 높은 형광 값이 발생했다. FHE와 Zn2+의 복합체화는 양자 수율(FHE, Φ = 0.016; FHEZn2+ 복합체, Φ= 0.34)이 20배 증가하면서 큰 스톡스 이동(Δλ = 198 nm, λex = 305 nm,λem = 503 nm)을 나타냈다. FLHE와 Zn2+의 복합체화는 양자 수율(FLHE, Φ = 0.007; FLHE-Zn2+ 복합체, Φ= 0.23)이 33배 증가하면서 큰 스톡스 이동(Δλ = 156nm, λex =350nm, λem = 506nm)을 나타냈다. 특히, 잠재적으로 경쟁하는 양이온과 음이온의 간섭 없이 녹색 형광으로 Zn2+ 이온을 검출하고 LOD가 각각 12.7nM과 31.8nM인 FHE와 FLHE는 높은 생체적합성을 나타냈다. FHE와 FLHE는 세포 내 Zn2+ 이온의 나노몰 검출에 성공적으로 사용되어 라이브 셀 이미징 적용 가능성을 보여준다. 또한 FHE와 FLHE는 환경 샘플에서 Zn2+를 검출하는 데 있어서 유망한 결과를 보여준다. 이러한 결과는 살아있는 세포와 강물, 수돗물과 같은 환경 샘플에서 Zn2+ 검출에 대한 잠재적인 응용을 검증한다. 또한, 물 속의 Zn2+ 이온과 유기 용매의 수분 함량을 정량화하기 위해 FLHE 기반 종이 스트립(FLHE-PS) 분석법이 개발되었다. FLHE-PS를 사용하면 LOD가 63.2nM인 수용액에서 Zn2+를 검출할 수 있으며 LOD가 0.14%인 아세토니트릴의 물에서 정량화할 수 있습니다. 이러한 결과는 FLHE가 살아있는 세포와 환경 시료에서 Zn2+를 검출하고 유기 용매에서 물의 존재를 검출하는 데 높은 적용 가능성을 가지고 있음을 나타낸다.키워드: 형광; 벤지미다졸; in vitro; 항암활성; 아연.
Recently, interest in targeting microtubule cytoskeletons to develop anticancer drugs is increasing. In this study, we developed a series of benzimidazole derivatives targeting intracellular and extracellular microtubule networks, especially targeting the binding site on the surface of β-tubulin. Compounds BE7-7 and O7-7 showed impressive anti-proliferative activity across various tested cell lines, showing selectivity index of 15.74 and 33.15, respectively. For cancer cell lines, BE7-7 achieved an IC50 value of 1.42 to 7.8 μM, while O7-7 showed an IC50 value of 1.13 to 8 μM. BE7-7 and O7-7 showed significant inhibition against wound closure, with the wound closure rate decreasing from 37-38% at 0 μM to 0.43% and less than 0% at 20 μM. Surprisingly, the colony formation reduction rate was remarkable at 95.8% for BE7-7 and 99.97% for O7-7. These results indicate that BE7-7 and O7-7 have the ability to interfere with cancer cell migration and have high potential to inhibit cloning. Cell cycle interruption led to cell cycle interruption at the G2/M stage, indicating that BE7-7 and O7-7 exhibit anticancer activity through the de-stabilization of microtubules. Additionally, the effect of inhibiting the tubulin network within cancer cells was observed by immunofluorescence assay. Overall, our study demonstrated the potential of synthesized benzimidazole analogs as novel and selective anticancer agents.We also developed two imaging probes, FHE and FLHE, which are Schiff base derivatives of the natural compound eugenol. Both of these derivatives showed high specificity for Zn2+ detection in biological and environmental samples. FHE and FLHE alone showed low fluorescence signal due to intramolecular charge transfer (ICT). However, titration with Zn2+ ions in HEPES buffer (pH = 7.4, 50% ACN, v/v) resulted in the formation of highly specific FHE–Zn2+ (Ka = 1.1 × 104 M-1) and FLHE–Zn2+ (Ka = 5.33 × 103 M-1) complexes. The complexation of probes with Zn2+ disrupts the ICT and subsequently results in chelation-enhanced fluorescence (CHEF) increment. The complexation of FHE with Zn2+ demonstrated a large stokes shift (∆λ = 198 nm, λex = 305 nm, λem = 503 nm) with 20-folds increase in the quantum yield (FHE, Φ = 0.016; FHE–Zn2+ complex, Φ = 0.34). The complexation of FLHE with Zn2+ demonstrated a large stokes shift (∆λ = 156 nm, λex = 350 nm, λem = 506 nm) with 33-folds increase in the quantum yield (FLHE, Φ = 0.007; FLHE–Zn2+ complex, Φ = 0.23). In particular, FHE and FLHE detected Zn2+ ions with green fluorescence with the LODs of 12.7 nM and 31.8 nM, respectively. Moreover, both of these probes did not show any interference from potentially competing cations and anions. FHE and FLHE have been successfully used for the nanomolar detection of intracellular Zn2+ ions, indicating its live-cell imaging applicability. Further, FHE and FLHE also showed promising results in detecting Zn2+ in the environmental samples. These results validate their potential applications for Zn2+ detection in living cells and environmental samples (river water and tap water). Additionally, a FLHE-based paper strip (FLHE-PS) assay was developed to quantify Zn2+ ions in water. FLHE-PS allows detection of Zn2+ in aqueous solutions with an LOD of 63.2 nM. The FLHE-PS also allows to detect and quantify water content in organic solvent (acetonitrile) with the LOD of 0.14%. These results indicate that FLHE has high applicability in detecting the presence of water in organic solvents.
