미량원소 아연은 골 형성과 발달에 관여하며 결핍시 성장 지연을 일으키는 것으로 알려져 있다. 아연이 골 형성에 미치는 기능에 대해서in vivo 연구에서는 잘 알려져 있으나, 세포수준에서의 연구는 미흡하다. 본 연구에서는 아연의 골 형성에 대한 기능을 조골세포인 MC3T3-E1 cell을 모델로 하여 두 가지 세부연구를 행하였다. 첫 번째 연구는 아연이 결핍시 조골세포 bone nodule 형성을 하는 세포외 무기질화가 저하되는지를 알아보았고, 두 번째 연구는 조골세포가 아연에 의해서 호르몬 leptin세포신호전달을 조절하는지를 JAK2-STAT3 경로를 중심으로 규명하였다. 첫 번째 연구에서, 두 가지 조골세포 subclone (subclone 4 및 negative control 로서 subclone 24을 사용하여 control (normal osteogenic medium), Zn-deficeincy (1 uM, Zn-)과 Zn-adequacy (15 uM, Zn+)에서 20일간의 골형성기간 동안 배양하였다. 조골세포 MC3T3-E1 subclone 4 는 무기질화를 촉진 시키는 특성을 지닌 세포라인이고 MC3T3-E1 subclone 24 는 무기질화를 감소시키는 특성을 지닌 세포이다. 두 subclone 모두에서 아연 결핍 시 Alizarin Red S staining과 von-Kossa staining 방법을 통해 세포외 기질의 Ca deposits을 측정을 통한 기질 무기질화 (extracellular matrix mineralization)를 측정한 결과, 아연 결핍하에서 조골세포외 무기질화는 두 subclone 모두에서 감소하였다. 또한 아연 결핍시, 세포외기질 bone-marker 유전자 및 단백질 (alkaline phosphatase, osteocalcin, osteopontin)의 발현이 저하되었다. 본 연구결과는 조골세포에서 아연이 결핍 시 세포외기질 Ca 축적과 bone marker protein의 발현 감소를 통해서 세포외 무기질화가 저하되고, 따라서 조골세포에 의한 골형성 기능도 저하되는 것으로 해석된다. 두 번째 연구에서는 조골세포에서 아연이 호르몬 leptin을 합성, 분비하는지, 또한 아연이 조골세포에서 leptin의 신호전달 경로를 조절하는지에 대해서 규명하였다. 이는 일반적으로 아연이 leptin의 발현을 조절하고, 근래에 leptin 조절이 골형성에 관여하는 것으로 보고 되고 있으나, 아직 이에 대해서는 논쟁이 되고 있기 때문이다. 즉, 두 번째 연구에서는 아연이 leptin 발현과 JAK2/STAT3 경로를 통한 leptin signaling을 조절하는지, 그리고 조골세포에서 이와 같은 조절을 통해서 골형성에 어떻게 관련이 있는지를 뒷받침하기 위해서 실험하였다. 조골세포 MC3T3-E1 cell을 OSM control, Zn-, Zn+에서 24 시간 동안 배양한 다음, leptin 분비와 leptin, leptin receptor (OB-Rb), leptin signaling 분자물인 JAK2와 STAT3 및 이들의 인산화 분자물의 유전자와 단백질 발현을 측정하였다. 또한 아연 결핍하에서의 세포사을 TEM (transmission electron microscopy)를 통해서 측정하였다. 아연 결핍 시 조골세포 MC3T3-E1 cell에서 leptin 유전자와 단백질 발현이 증가하였고, 세포 외 leptin 분비가 증가되는 경향을 나타내었다. 세포 내 leptin signaling의 leptin receptor (OB-Rb) 유전자와 단백질 발현 또한 아연 결핍 시에 증가되었는데 아마도 이는 증가된 leptin 분비로 인한 것으로 해석된다. 또한 아연 결핍시, leptin signaling 분자물인 인산화된STAT3 (pSTAT3)가 핵 내에서의 발현이 증가한 것으로 나타났다. TEM에 의한 이미지는 아연 결핍시 세포사멸 증상을 일부 나타내었다. 본 연구결과는 아연 결핍 시 조골세포 MC3T3-E1 세포에서는 leptin의 합성 및 분비가 증가되는 양상을 보이며, 따라서 증가된 leptin분비가 JAK2-STAT3 경로를 활성화시키는 것으로 해석된다. 일반적으로 STAT3가 활성화 되면 세포 생존 유전자들의 전사가 축진 되는 것으로 알려져 있으므로, 본 연구의 결과는 아연 결핍시 활성화된 leptin JAK2-STAT3 경로는 아마도 아연 결핍시 나타나는 세포사멸을 막기 위한 세포 조절 현상일 것으로 생각되며, 이에 대한 후속 연수가 필요하다. 결론적으로 아연의 조골세포에서의 골형성에 관한 조절기능을 규명한 본 연구는, 미량원소 아연은 세포외 기질Ca 축적과 골형성 지표 단백질 (ALP, OC, OPN) 발현 증가를 통하여 세포외기질 무기질화를 촉진시키고, 세포사멸이 유도되는 아연 결핍하에서 JAK2-STAT3 를 통한 leptin signal를 촉진시켜 아연 결핍하에서의 세포사멸을 줄이기 위한 항상성(homeostasis)을 유지하려고 하는 것으로 해석된다. 본 연구결과는 아연이 조골세포에서 골형성을 촉진하는 기능을 규명하였고, 이는 골다공증과 같은 골 관련 질병을 예방 하는데에 기초 자료로 쓰일 것이다.
Zinc deficiency is associated with growth retardation which implies zinc involvement on bone formation. The role of zinc on bone formation is well documented on the stimulatory effect in vivo study; however, the mechanism at cellular level is not clearly understood, yet. To clarify the role of zinc for bone formation by osteoblasts, two studies were examined: The first study investigated whether zinc deficiency decreases extracellular matrix (ECM) mineralization in osteoblastic MC3T3-E1 cells, since Ca deposits and bone marker protein accumulation at ECM are the major parts of ECM mineralization process for bone formation. The second study investigated whether zinc modulates leptin signaling via JAK2-STAT3 pathway in MC3T3-E1 cells, and how that modulation would be relevant to bone formation, since zinc is a regulator for leptin expression and leptin is of interest as an osteogenic regulator as autocrine way. At the first study, it was investigated whether zinc deficiency would decrease ECM mineralization during bone formation. Osteoblastic MC3T3-E1 subclone 4 (mineralizing one), along with MC3T3-E1 subclones 24 (nonmineralizing one) which was characterized as to less mineralize an extracellular matrix, was cultured in control (normal osteogenic media, OSM), Zn-deficiency (1 μM, Zn-) and Zn-adequacy (15 μM, Zn+) with the addition of 5 μM TPEN as intracellular zinc chelator up to 20 days of bone formation period. Ca deposits assessed by Alizarin Red S and von-Kossa staining showed ECM mineralization is lower in zinc-deficiency in both subclones. The expression of ECM bone-marker proteins (ALP, OC, OPN) which is necessary for ECM mineral deposition was lower in zinc-deficient osteoblasts. The study results suggested that zinc deficiency decreases ECM mineralization via decreased ECM Ca deposits and bone marker protein expression in osteoblast, which may in turn inhibit bone formation by osteoblasts. The first study showed a positive role of zinc for bone formation through ECM mineralization in osteoblasts, and zinc has been shown to mediate leptin expression which makes leptin as a candidate molecule linking zinc with bone formation. Therefore, at the second study, it was investigated whether zinc modulates leptin expression and leptin signaling through JAK2-STAT3 pathway, and how that modulation would be relevant to bone formation in osteoblasts. Osteoblastic MC3T3-E1 cells were cultured in OSM, Zn- and Zn+ as described above for 24 h and gene and protein expression of molecules for leptin signaling, such as leptin, leptin receptor (OB-Rb), JAK2 and STAT3, were measured. Cell apoptosis under zinc deficiency was also assessed by transmission electron microscopy. Zinc deficiency increased leptin gene/protein expression and extracellular leptin secretion. Leptin-receptor (OB-Rb) gene/protein expressions for intracellular leptin signaling were also increased in zinc-deficiency, perhaps due to the increased leptin secretion. Protein and phosphorylated expression of cytosolic leptin signaling molecules (JAK2, pJAK2, STAT3 and pSTAT3) were increased and conveyed the phosphorylated STAT3 (pSTAT) in the nucleus in zinc-deficiency. Even under the increase of the activated STAT3 in the nucleus, which may stimulate the transcription of cell survival genes, cell apoptosis was observed under zinc deficiency. The study results suggest that zinc deficiency activates leptin signaling through JAK2-STAT3 pathway in mouse osteoblastic MC3T3-E1 cells and this could be considered as a compensatory regulation to increase cell survival to resist to cell apoptosis which was induced under zinc deficiency. Taken all together, it can be suggested that zinc can increase bone formation through increasing ECM mineralization via Ca deposits and bone marker protein expression, and regulating as compensatory role to prevent cell apoptosis which was caused under zinc deficiency in osteoblastic MC3T3-E1 cells. Potential role of zinc for bone formation by osteoblasts can be a strategy for preventing bone disease such as osteoporosis.
