PURPOS : This study was to examine the effect of different exercise intensity and diet on the expression of the metabolism related factors in skeletal muscles of T2DM rats.METHODS : Type 2 diabetes was induced in rats by a combination of low dose streptozotocin(STZ; 40 mg/kg) and feeding of a high fat diet. The rats had trained by low intensity(40% VOmax), high intensity(80% VOmax) exercise on the treadmill for 8 weeks. Then the rat soleus(SOL) and extensor digitorum longus(EDL) were taken out. The western blotting was performed for the expression of AMPK, p-AMPK, PGC-1α, and GLUT-4 proteins.RESULTS : The expression of AMPK, p-AMPK, PGC-1α, and GLUT-4 proteins in EDL and Soleus muscle were higher in both training groups compared to the non-training groups. The AMPK, p-AMPK, PGC-1α, and GLUT-4 proteins significantly in normal diet and endurance training than in high fat diet and endurance training. The AMPK, p-AMPK, PGC-1α, and GLUT-4 proteins expressions in normal diet and endurance training was highest. CONCLUSIONS : This study can be concluded that the expression of AMPK, PGC-1α and GLUT-4 proteins was differed with exercise intensity, diet and the type of muscle. These results indicated that the endurance training improved the insulin sensitivity according to the exercise intensity and diet function in T2DM rats.
