Pyropia yezoensis 는 홍조류에서 가장 중요한 경제적 작물 중 하나다.Porphyran 은 식용 Pyropia yezoensis 의 풍부한 활성 성분으로 항염, 혈당 저하, 지질혈증 완화 및 기타 약리 효과를 가지고 있다. 따라서, porphyran 은 기능성 식품 및 의약품을 개발할 수 있는 잠재력을 갖고 있다. 현재, 고당류 식사로 인한 대사 장애 및 질병이 어린이, 청소년 및 성인에게 만연하다. 그러나 대사 장애 및 그 질병에 대한 수술 및 약물치료는 부작용이 많고 비용이 많이 들어 안전하고 효과적인 치료 전략이 필요하다. 천연 다당류 프리바이오틱스는 장내 미생물총을 조절함으로써 대사 장애 및 그 질병의 조정 및 치료를 위한 신종 치료법이다. 본 연구의 목적은 항대사활성을 갖는 P. yesoensis 에서 유래한 porphyran 의 기능성 확인 과 그 기작을 연구하는 것이다. P. yesoensis 으로부터 porphyran 은 열수추출법으로 얻었고, 저분자량 porphyran 은 감마선 조사를 통해 제조했으며, porphyran 및 그 유도체들의 물리화학적 특성을 분석했다. 동시에, 시험관 및 생체 내에서 체계적으로 porphyran 의 생물학적 활성을 연구했다.제1장. 발표된 기사를 검토하기 위해 Web of Science 의 데이터에 키워드“고당류 식사”, “대사 장애 및 질병”, “다당류” 및 “파래”를 검색했다. 고당류 식사에 의해 유발되는 유행성 대사 질병의 배경과 치료 방법이 소개됐다. 동시에 홍조류 porphyran 의 연구 현황과 인체 건강에 미치는 영향을 요약하였다. 현재까지 P. yesoensis 에서 유래한 porphyran 의 고당 유발 대사 장애에 대한 조정 효과 및 기전에 관한 연구는 보고되어 있지 않다.제2장. P. yezoensis 에서 열수추출법으로 porphyran (PYP) 을 추출했다. 감마선 조사를 통해 두 가지로 분해된 porphyran 를 만들어 PYP-20 과 PYP-50 이라고 명명했다. 비색, 분광 (푸리에 변환 적외선 분광 및 원편광 이색성) 및크로마토그래피 (PMP-고성능 액체 크로마토그래피 및 고성능 겔 투과 크로마토그래피) 를 통해 PYP, PYP-20, PYP-50 의 물리화학적 및 구조적 특성을 연구했다. 그 결과 PYP 가 수용성 삼중나선 황화 다당류라는 것이 밝혀졌다. PYP 는 20 kGy 및 50 kGy 의 감마선 조사에 의해 분해되어 각각 두 개의 저분자량 유도체 PYP-20 과 PYP-50 을 제공했다. PYP, PYP-20, PYP-50 의 구성에는 큰 차이가 없으며 모두 황산염, 갈락토오스 (Gal), 자일로스 (Xyl), 아라비노오스 (Ara), 그리고 미량 단백질로 구성되어 있다. 이들은 서로 다른 분자량을 가지고 있어, PYP 와 두 개의 분해된 porphyran 사이의 용액 구조도 차이가 난다. 이러한 분석을 통해 PYP, PYP-20, PYP-50 사이의 가장 큰 차이가 분자량임을 확인하였고, 그들의 생물학적 활성 연구를 진행하였다. 제3장. PYP, PYP-20, PYP-50 이 대사 장애를 개선하는 능력이 있는지 조사하기 위해 서로 다른 세포주에서 당 처리에 의해 유도된 비정상적인 지질대사를 시험관 내 모델로 사용했다. 그 결과 당에 의한 대사 장애는 HepG2 와 Caco-2 세포에서 당 농도에 의존하는 것으로 나타났으며, 특히 고당 농도로 인한 지질 축적이 가장 뚜렷하게 나타났다. 또한, 고농도의 PYP 보충은 PYP-20 및PYP-50 에 비해 고당류 (HS) 그룹의 비정상적인 대사에 더 좋은 억제 활성을 가지고 있어 이는 분자량 및 농도에 의한 것일 수 있음을 시사한다. 따라서, 고농도의 PYP 를 선택하고 oil red O staining 과 RT-qPCR 을 통해 PYP 가 지질 축적을 감소시키는 잠재적 기작을 분석했다. PYP 는 고당으로 유도된 epG2 와 Caco-2 세포에 지질 방울이 축적되는 것을 현저히 감소시켰다. 더욱이, PYP 는 Srebp 와 Fas (지방생성에 관여하는 유전자) 의 발현을 억제하고 이들 세포에서 PPARα 와 Cpt1 (지방산 β-산화에 관여하는 유전자)의 발현을 증가시켰다. 이는PYP 가 지질 합성을 감소시키고 지방산 β-산화를 증가시켜 대사 이상을 개선함으로써 지질 저하 활성을 발휘한다는 것을 보여준다.제4장. PYP 가 체내에서도 대사 장애를 완화시키는 활성이 있는지 확인하기 위해, Drosophila 유충에게 당을 먹여 유도된 대사 이상을 체내 모델로 사용했다. 그 결과는 당에 의한 대사 장애와 Drosophila 유충의 발달 지연이 당 농도와 긍정적인 상관관계가 있다는 것을 보여주었고, 그 중에서도 고당류 (HS) 그룹이 가장 두드러졌다. 따라서, PYP 활성의 기전을 더 탐구하기 위해 고당류 그룹을 모델로 선택했다. 고농도의 PYP 가 HS 그룹에서 트리글리세라이드 및 순환 당 함량을 유의미하게 억제하였으므로, 25 mg/mL PYP 를 선택하여 대사 이상에 대한 영향을 추가로 조사했다. 그러나 PYP 는 장내 미생물총을 제거하기 위해 혼합 항생제를 사용한 HS 그룹 유충의 트리글리세라이드와 순환 당 함량에는 영향을 미치지 않았다. 이는 PYP 의 활성이 장내 미생물총과 관련이 있음을 나타낸다. 또한, 유충 내 일부 대사 관련 유전자를 분석했다. PYP 보충은 유충에서 Srebp 및 Fas (지방 생성에 관여하는 유전자)의 발현을 억제하고 Fabp 및 Acox57D-d(지방산 β-산화에 관여하는 유전자)의 발현을 증가시켜, 시험관 내 비정상적인 지질 대사를 완화하는 것과 일치했다. 또한, PYP 는 HS 그룹의 번데기 및 성인단계에서 비정상적인 대사를 억제했다. PYP 보충이 유충에서 성충으로 발달 결함을 교정한 것으로 나타났다. 요컨대, PYP 는 대사 장애를 예방하고 치료하기 위한 프리바이오틱으로 사용될 수 있다.제5장. 고당 (HS) 에 의해 유도되는 대사 장애를 개선하는 PYP 의 잠재적 기작을 탐구하기 위해, 각 그룹에서 유충의 발달 말기에 있는 삼령 유충을 선택했다. 고당류를 섭취한 Drosophila 유충의 장내 미생물총에 대한 PYP 의 영향이 16S rDNA 염기서열 분석으로 연구되었고 주요 변화 박테리아가 확인됐다. 생화학적 분석과 RT-qPCR 분석에 따르면 HS 식이요법은 16S rDNA 분석에 의해 파악된 유해 미생물총의 분포 뿐만 아니라 Drosophila 유충의 트리글리세라이드 및 순환 당 함량 (대사 이상) 을 증가시켰다. PYP 보충은 대사 이상을 현저히 감소시키고 HS 그룹의 Escherichia-Shigella 및 Fusobacterium 과 같은 유해 박테리아의 풍부함을 감소시키는 한편, Bacillus 및 Akkermansia 와 같은 유익한 박테리아의 풍부함을 증가시켰다. 기능적 예측은 PYP 가 HS 유도 대사 장애를 줄이기 위해 대사 경로에 영향을 미칠 수 있음을 시사했으며, 대사 매개변수의 분석은 이러한 결과와 일치했다. 또한, PYP 는 정상 그룹과 비교했을 때 항생제가 제거된 혼합 장내 미생물총 유충의 트리글리세라이드 및 순환 당 함량에는 아무런 영향을 미치지 않는다는 사실도 알아냈다. 이러한 데이터는 PYP 가 장내 미생물총 및 주요 박테리아 속의 변화를 조절함으로써 HS 에 의해 유도된 대사 장애를 완화할 수 있음을 나타낸다.이상의 결과는 PYP 가 천연 홍조류 유래 프리바이오틱으로, 유익한 박테리아는 증가시키고 유해 박테리아는 감소시켜 성충에 대한 Drosophila 유충의 비정상적인 발육을 완화시켜 대사 장애를 개선하는 활성을 나타낸다.종합하면, P. yezoensis porphyran (PYP) 은 건강 증진을 위한 해양 기능성 식품 및천연 다당류 의약품으로 활용될 수 있다.
Pyropia yezoensis is one of the most important economic crops in red algae. Porphyran,an abundant active ingredient in edible Pyropia yezoensis, has pharmacological effects suchas anti-inflammatory, hypoglycemic, and hypolipidemic. Therefore, porphyran has potentialfor exploitation in functional foods and medicine. Currently, metabolic disorder and diseasescaused by a high-sugar diet are prevalent in children, adolescents, and adults. However,surgical and pharmacological treatments for metabolic disorder and their diseases have manyside effects and are expensive, hence the need for safe and effective treatment strategies.Natural polysaccharide prebiotics is a novel therapy to intervene and treat metabolic disorderand diseases by modulating the gut microbiota. In this study, the aim was to discover P.yezoensis porphyran with anti-metabolism activity. P. yezoensis porphyran was obtained byhot water extraction, and low molecular weight porphyran was prepared by gamma irradiation,while the physicochemical properties of porphyran and its derivatives were analyzed.Simultaneously, the biological activity of porphyrans was systematically investigated in vitroand in vivo.Chapter Ⅰ. Data onto Web of Science were searched using the keywords “high-sugardiet”, “metabolic disorder and diseases”, “polysaccharides”, and “laver”, and publishedarticles were reviewed. The background and treatment of epidemic metabolic diseases inducedby a high-sugar diet were introduced. Simultaneously, the research status of red algaeporphyran and its influence on human health was summarized. Currently, the interventionaleffects and its underlying mechanism of P. yezoensis porphyran on high-sugar-inducedmetabolic disorder have not been concerned.Chapter Ⅱ. Porphyran (PYP) was extracted from P. yezoensis by the hot water extractionmethod. Two degraded porphyrans, named PYP-20 and PYP-50, were prepared by gammairradiation. The physicochemical and structural properties of PYP, PYP-20, and PYP-50 wereinvestigated by colorimetric, spectroscopic (Fourier transform infrared spectroscopy andcircular dichroism), and chromatographic (PMP-high performance liquid chromatography andhigh-performance gel permeation chromatography) methods. The results showed that PYP isa water-soluble triple helix sulfated heteropolysaccharide. PYP was degraded by gammairradiation at 20 kGy and 50 kGy, giving two low molecular weight derivatives PYP-20 andPYP-50, respectively. There is no obvious difference in the composition of PYP, PYP-20, andPYP-50, all being composed of sulfate, galactose (Gal), xylose (Xyl), arabinose (Ara), andtrace protein. They have significantly different molecular weights, which also leads todifferences in solution conformation between PYP and the two degraded porphyrans. Theseanalyses confirmed that the most significant difference among PYP, PYP-20, and PYP-50 wasthe molecular weight, and further explore their biological activity.Chapter Ⅲ. To investigate whether PYP, PYP-20, and PYP-50 have the ability toameliorate metabolic disorder, the abnormal lipid metabolism induced by sugar treatment indifferent cell lines was used as an in vitro model. The results showed that the sugar-inducedmetabolic disorder showed a sugar concentration-dependent in HepG2 and Caco-2 cells,especially the lipid accumulation induced by high sugar concentration was the most significant.Furthermore, a high concentration of PYP supplementation showed better inhibitory activityagainst abnormal metabolism in the high-sugar (HS) group compared to PYP-20 and PYP-50,suggesting that this may be influenced by molecular weight and concentration. Therefore, ahigh concentration of PYP was selected and the potential mechanism by which PYP reduceslipid accumulation was analyzed by oil red O staining and RT-qPCR. PYP significantlyreduced the accumulation of lipid droplets in high sugar-induced HepG2 and Caco-2 cells.Furthermore, PYP inhibited the expression of Srebp and Fas (genes involved in lipogenesis)and increased the expression of PPARɑ and Cpt1 (genes involved in fatty acid β-oxidation) inthese cells. This suggests that PYP exerts lipid-lowering activity by reducing lipid synthesisand increasing fatty acid β-oxidation to improve metabolic abnormalities.Chapter Ⅳ. To verify whether PYP also has the activity to alleviate metabolic disorderin vivo, the metabolic abnormalities induced by sugar feeding to Drosophila larvae were usedas an in vivo model. The results showed that sugar-induced metabolic disorder anddevelopmental delay of Drosophila larvae were positively correlated with sugar concentration,especially in the high-sugar (HS) group. Therefore, the high-sugar group was selected as amodel to further explore the mechanism of PYP activity. A high concentration of PYPsignificantly suppressed triglyceride and circulating sugar contents in the HS group, therefore,25 mg/mL PYP was selected to further investigate the effects on metabolic abnormalities.However, PYP had no effect on triglyceride and circulating sugar contents of larvae in the HSgroup that used mixed antibiotics to remove the gut microbiota. This indicates that the activityof PYP is related to the gut microbiota. In addition, some metabolic-related genes in larvaewere analyzed. PYP supplementation suppressed the expression of Srebp and Fas (genesinvolved in lipogenesis) and increased the expression of Fabp and Acox57D-d (genes involvedin fatty acid β-oxidation) in larvae, which was consistent with alleviating abnormal lipidmetabolism in vitro. In addition, PYP also inhibited abnormal metabolism in the pupal andadult stages of the HS group. It was found that PYP supplementation corrected thedevelopmental defects from larvae to adults. In short, PYP may be developed as a prebioticfor the prevention and treatment of metabolic disorder.Chapter Ⅴ. To explore the potential mechanisms of PYP in improving the metabolicdisorder induced by high sugar (HS), third-instar larvae at the end of larval development wereselected from each group. The effect of PYP on the gut microbiota of Drosophila larvae fedwith high sugar was studied by 16S rDNA sequencing and key change bacteria were identified.Biochemical and RT-qPCR analysis showed that HS diet-feeding increased triglyceride andcirculating sugar contents (metabolic abnormalities) in Drosophila larvae, as well as theabundance of harmful microbiota as determined by 16S rDNA analysis. PYP supplementationsignificantly reduced metabolic abnormalities and decreased the harmful bacteria abundance,such as Escherichia-Shigella and Fusobacterium in the HS group while increased theabundance of beneficial bacteria, such as Bacillus and Akkermansia. Functional predictionssuggested that PYP might affect the metabolism pathway to reduce HS-induced metabolicdisorders, and the analysis of metabolic parameters was consistent with these consequences.Furthermore, we also found that PYP had no effect on triglyceride and circulating sugarcontents in larvae of mixed antibiotic-removed gut microbiota compared to the normal group.These data indicate that PYP can alleviate the metabolic disorders induced by HS by regulatingthe changes in the gut microbiota and key bacterial genera.The above results indicate that PYP is a natural red algae-derived prebiotic, whichexhibits the activity of improving metabolic disorder by increasing the beneficial bacterial andreducing the harmful bacterial, thereby alleviating abnormal development of Drosophilalarvae to adults. Taken together, it is recommended that P. yezoensis porphyran (PYP) bedeveloped as a marine functional food and natural polysaccharide medicine for promotinghealth.
