Aging is a progressive disease with weakness of physiological functions. The comprehensive metabolomics study of aging was not fully performed, although the molecular inflammation hypothesis related to oxidative stress and molecular inflammation has been established. Thus, in this study, metabolomic analysis of amino acids (AAs), organic acids (OAs), and fatty acids (FAs) in brain tissues from young rats and old rats was performed by gas chromatography-tandem mass spectrometry (GC-MS/MS). In the result, the 32 total metabolites of 22 AAs, nine OAs, and one FA in the cortex, the 43 total metabolites of 21 AAs, 10 OAs, and 12 FAs in the cerebellum, the 45 total metabolites of 22 AAs, 10 OAs, and 13 FAs in the hypothalamus, the 30 total metabolites of 18 AAs, nine OAs, and three FAs in the hippocampus of young and old groups were determined. Their concentration values were normalized to the corresponding mean values of young group, which were plotted as star graphs. These were characteristic and readily distinguishable of the difference for AAs, OAs, and FAs in each group. In the profile analysis, 10 metabolites (alanine, valine, leucine, isoleucine, threonine, serine, proline, phenylalanine, tyrosine, and lactic acid) in the cortex, 14 metabolites (alanine, glycine, valine, leucine, isoleucine, threonine, serine, proline, methionine, phenylalanine, cysteine, 3-hydroxypropionic acid, eicosadienoic acid, erucic acid) in the cerebellum, seven metabolites (glycine, β-alanine, valine, proline, γ-aminobutyric acid, cysteine, aspartic acid) in the hypothalamus, and acetoacetic acid in the hippocampus were evaluated as major metabolites with aging, which were significantly altered and obtained high VIP scores (>1.0) in OPLS-DA. In addition, the clustering of each group in all brain tissues was completely separated in OPLS-DA. Although statistically significant changes of AAs, OAs, and FAs in each brain tissue was different, these explained mitochondrial dysfunction and inflammation as agingrelated metabolism. Especially, mTORC1-related BCAA metabolism and inflammation-related metabolism were commonly altered in all brain tissues, which were significantly elevated in the cortex, cerebellum, and hypothalamus except for hippocampus. And, acetoacetic acid as a ketone body in the hippocampus was significantly elevated in old group compared to young group. These findings may explain altered aging energy metabolism, aging-related mitochondrial dysfunction, and increased inflammation by ROS accumulation and oxidative stress. Also, the AAs as neurotransmitters were significantly mostly altered in all brain tissue. These results may explain for imbalance of neurotransmitters in brain tissues with aging. In addition, three metabolites (leucine, tyrosine, and lactic acid) related metabolism were altered in cortex may explain the change for cortex function related to cognitive decline. Thus, in this study, metabolic changes including increased inflammation, altered energy metabolism related mitochondrial dysfunction, and imbalance of neurotransmitters in brain tissues with aging may be considered related to aging.In addition, the database of AA, OA, and FA profiles and metabolic patterns in brain tissues (cortex, cerebellum, hypothalamus, and hippocampus) of young and old groups were constructed by metabolite profiling and pattern analysis. Therefore, the present result will be useful for detection of potential biomarkers and monitoring of metabolic change related with aging in brain tissues.
노화는 생리적 기능이 약화되는 진행성 질환이다. 비록 노화에 대한 산화적스트레스와 염증 관련한 분자 염증 가설이 확립되었지만, 체계적인 대사체학연구는 거의 수행되지 않았다. 그러므로, 본 연구에서는 젊은 쥐와 노화 쥐의 뇌조직에서 기체 크로마토그래피-텐덤 질량분석법을 활용한 아미노산, 유기산, 지방산에 대한 대사체학 연구를 수행하였다 그 결과, 대뇌피질에서 32 종대사체 (아미노산 22 종, 유기산 9 종, 지방산 1 종), 소뇌에서 43 종 대사체(아미노산 21 종, 유기산 10 종, 지방산 12 종), 시상하부에서 45 종 대사체(아미노산 22 종, 유기산 10 종, 지방산 13 종), 그리고 해마에서 30 종의대사체 (아미노산 18 종, 유기산 9 종, 지방산 3 종)를 확인하고 정량분석을수행하였다. 젊은 쥐와 노화 쥐의 뇌 조직에서 정량한 대사체들은 젊은 쥐그룹의 평균값으로 정규화한 다음 스타그래프를 작성하였다. 스타그래프들은특징적이었고 각 그룹에 대한 아미노산, 유기산 및 지방산의 차이를 쉽게확인할 수 있었다. 대사체 프로파일에 대한 통계 분석 결과, 대뇌피질에서는 알라닌, 발린, 류신, 아이소류신, 트레오닌, 세린, 프롤린, 페닐알라닌, 티로신, 젖산을 포함한 10 종, 소뇌에서는 알라닌, 글리신, 발린, 류신, 아이소류신, 트레오닌, 세린, 프롤린, 메티오닌, 페닐알라닌, 시스테인, 3-하이드록시프로피온산, 에이코사디에노산, 에루크산을 포함한 14 종, 시상하부에서는 글리신, 베타-알라닌, 발린, 프롤린, 감마-아미노뷰티르산, 시스테인, 아스파르트산을 포함한 7 종, 그리고 해마에서는 아세토아세트산이통계적으로 유의적인 변화를 나타내었다. 그리고 이들 대사체들은 OPLSDA 에서 젊은 쥐와 노화 쥐 그룹에 대한 높은 판별력 (VIP scores >1.0) 을나타내는 대사체들로 평가되었으며, 모든 뇌조직에서 두 그룹은 완전히분리됨을 확인하였다. 비록, 각 뇌조직에서 아미노산, 유기산 및 지방산의유의적인 변화는 달랐지만, 변화된 대사체들은 미토콘드리아 기능 장애 및염증으로 노화 관련 대사와 연관되어 있음을 알 수 있었다. 특히, mTORC1 관련한 분지쇄아미노산 대사 및 염증 관련 대사는 모든 뇌 조직에서 공통적으로변화하였으며 해마를 제외하고 대뇌피질, 소뇌 및 시상하부에서는 유의적으로증가하였다 그리고 해마에서 케톤체인 아세토아세트산은 ?은 쥐에 비해 노화쥐에서 유의하게 증가하였다. 이러한 결과는 활성산소종의 축적과 산화적스트레스에 의해 증가되는 염증과 노화 관련 미토콘드리아 기능 장애 및노화관련 에너지 대사 변화를 설명할 수 있다. 또한 신경 전달 물질로서아미노산들은 모든 뇌 조직에서 유의하게 변화하였다. 이러한 결과는 노화에따른 뇌 조직의 신경 전달 물질의 불균형을 설명할 수 있다. 그리고 대뇌피질에서 변화한 세 종의 대사체 (류신, 티로신, 젖산)는 인지 능력과 관련된기능 저하로 설명할 수 있다. 그러므로, 본 연구에서, 노화에서의 염증 증가, 미토콘드리아 기능 장애와 관련된 에너지 대사 변화, 뇌 조직의 신경 전달 물질불균형은 노화와 관련된 변화로 사료된다. 그리고, 대사체 프로파일과 패턴분석을 통해 젊은 쥐와 노화 쥐의 뇌 조직 (대뇌피질, 소뇌, 시상하부,해마)에서의 아미노산, 유기산 및 지방산 프로파일과 대사 패턴에 대한 데이터베이스를 구축하였다. 그러므로, 본 연구 결과는 뇌 조직의 노화와 관련된 대사변화 모니터링 및 잠재적 바이오 마커 탐색에 유용할 것으로 사료된다.
