Norovirus (NoV), hepatitis A virus (HAV), and hepatitis E virus (HEV) are representatives among the most common foodborne viruses. Consumption of food infected with these viruses causes chills, vomiting, and diarrhea, which in turn causes great social and economic losses. Therefore, it is necessary to build a system to detect these foodborne viruses. There have been several methods for detecting viruses, including conventional polymerase chain reaction (conventional PCR), real-time quantitative polymerase chain reaction (RT-qPCR), and enzyme-linked immunosorbent assay (ELISA). However, these detection methods are limited by the need for expensive equipment, laboratory experts, and long reaction times, making field validation impossible. To overcome these limitations, this study developed fluorescence-based real-time loop-mediated isothermal amplification (RT-LAMP) assay for NoV, HAV, and HEV. In the case of the LAMP assay, false-positive reactions are frequent due to their high sensitivity. Although many factors contribute to false-positive, this study focused on the formation of a secondary structure of the primers to solve this problem. Herein, the developed sensor showed a low detection limit of 1.6 copies/µL for NoV, 3.9 copies/µL for HAV, and 31.6 copies/µL for HEV, respectively. Furthermore, it showed high specificity for each target compared to other types of viruses.
Norovirus (NoV), hepatitis A virus (HAV), hepatitis E virus (HEV)는 대표적인 식중독을 유발하는 바이러스 중 하나이다. 이러한 바이러스들에 감염된 음식을 섭취를 하게 되면 오한, 구토 및 설사를 일으키게 되며 결국에는 사회, 경제적인 큰 손실을 일으킨다. 그렇기 때문에 이러한 식중독 유발 바이러스들을 검출하는 시스템을 구축할 필요성이 대두되고 있다. 바이러스를 검출하는 방법에는 전통적인 중합효소연쇄반응 (conventional PCR), 실시간 중합효소연쇄반응 (RT-qPCR), 효소결합면역흡착분석법 (ELISA) 등 여러 가지가 있다. 그러나 이러한 검출 방법들은 고가의 장비, 실험 전문가 및 긴 반응 시간이 필요하여 현장 검증이 불가능하다는 한계가 있다. 이러한 한계를 보완하기 위해 본 연구에서는 형광 기반 실시간 루프 매개 등온 증폭법을 이용하여 NoV, HAV 그리고 HEV에 대한 각각의 특이적인 프라이머를 개발했다. 그러나 루프 매개 등온 증폭법의 경우, 높은 민감도로 인해 위양성 반응이 빈번하게 나타나며 이에 대한 원인으로는 여러 요인들이 있지만, 본 연구에서는 프라이머의 2차 구조 형성에 초점을 두어 이를 해결하는 연구를 진행하였다. 이렇게 개발된 센서는 NoV는 1.6 copies/µL, HAV는 3.9 copies/µL, HEV는 31.6 copies/µL의 낮은 검출한계를 드러냈다. 또한 다른 종류의 바이러스들과 비교하였을 때 각 타겟 물질에만 높은 특이성을 나타냈다.
