Ultrasound neuromodulation is a promising tool for depression disorder (DD) therapy. Research to date has focused on single-target stimulation such as prelimbic cortex (PrL) or dorsal raphe nucleus (DRN). However, the pathogenesis of DD involves multiple cerebral connected regions. A simultaneously simulating multiple regions associated with depression pathology is required to allow more efficient treatment. In this study, we investigate antidepressant effect of multi-target ultrasound neuromodulation of PrL and DRN. The effects were evaluated by liquid chromatography-mass spectrometry (LC-MS) and western blot (WB) to detect BDNF in PrL and 5-HT in DRN, respectively. The results indicated that simultaneously simulating PrL and DRN has a more pronounced antidepressant effect.
