Understanding of spatial and temporal pattern for the activation of resident cardiac stem cells in three-dimension pattern will be helpful for us to demonstrate the cellular and molecular mechanism of regeneration of infarcted myocardium and tailor novel strategy to cure myocardial infarction. In present study, we apply fluorescent image consecutive scanning combing with three-dimensional reconstruction to display the c-kit + and activating c-kit + -cardiac stem cells. The results of present study revealed that 3D-reconstruction technique is able to display and measure the c-kit + or activating c-kit + -cardiac stem cells in the 3D version. Using this unique technique, we are not only able to display the spatial relationship of labeled structures and cell-to-cell in free angle with 3D-dimension, but also able to compare the labeled structures quantitatively.
