研究聚乙二醇化重组人干扰素α-2b(PEG-IFN)和重组人干扰素α-2b(IFN)的体外抗乙型肝炎病毒作用.以HepG2.2.15为细胞模型,采用MTT法考察两种药物对细胞的毒性作用;分别以高、中、低剂量的PEG-IFN和IFN每隔一日反复作用细胞,连续8d,采用ELISA法测定细胞上清液中的HBsAg来考察两种药物对病毒的抑制作用;将相同剂量的PEG-IFN和IFN模拟临床给药频率作用细胞一周,即PEG-IFN给药1次,IFN给药3次,观察细胞HBsAg的分泌情况,评价药效.PEG-IFN和IFN的TC_(50)均>20000IU/mL,IC_(50)分别为559.88、478.71IU/mL;反复给药时,高、中、低3个剂量组的PEG-IFN对病毒的抑制作用略低于同剂量的IFN;模拟临床给药一周后发现,PEG-IFN与IFN对病毒的抑制作用无明显差异.PEG-IFN和IFN在体外均可抑制HepG2.2.15细胞乙肝病毒的表达.
To study the inhibitive activities of PEG-IFN and IFN against hepatitis B virus in vitro,HepG2.2.15 cell line was used as cell model.MTT assay was adopted to evaluate the toxicity of PEG-IFN and IFN on cell line.High,medium and low doses of PEG-IFN and IFN were added to the cell every other day for 8 days.ELISA was adopted to detect the level of HBsAg in supernatant.Same doses of PEG-IFN and IFN were added to the cell according to the frequency of clinical administration for one week,that is PEG-IFN was added for 1 time while IFN was added for 3 times,ELISA to determine the secretion of HBsAg.TC_(50) (PEG-IFN) > 20000IU/ mL,TC_(50) (IFN) > 20000IU/mL.IC_(50) (PEG-IFN)=559.88 IU/mL,IC_(50) (IFN)=478.71IU/mL.The inhiting ratio of PEG-IFN was a bit lower than that of IFN with the same doses.The inhibition of the both have no significant differences according to the frequency of clinical administration.PEG-IFN and IFN have a noticeable anti-HBV effect with low toxicity in vitro,which show fine applying prospeers.
