目的 研究Th17、Treg细胞检测在小儿EB病毒(EBV)感染中的临床意义.方法 回顾性选取2021年1月至2023年1月泸州市人民医院收治的60例EBV感染患儿作为研究组,同时选取同一时期入院进行体检的60名健康儿童作为对照组.采用流式细胞仪测定外周血T淋巴细胞内的Th17、Treg细胞,并采用酶联免疫吸附抗体双夹心法测定相关特征性细胞因子[转化生长因子-β(TGF-β)、白细胞介素17(IL-17)]水平;同时,采用RT-PCR法测定Th 17表达孤核受体(RORγt)mRNA、Treg表达叉头翼状螺旋转录因子(Foxp3)mRNA表达水平.比较两组患者的外周血Treg细胞表达率、Th17细胞表达率、TGF-β、IL-17水平与RORγt mRNA、Foxp3 mRNA相对表达水平.结果 研究组患儿外周血Th17细胞表达率为(1.39±0.42)%,明显高于对照组[(0.82±0.19)%],外周血Treg细胞表达率为(3.48 ±0.84)%,明显低于对照组[(6.07±1.25)%],差异均有统计学意义(P<0.05).研究组患儿外周血RORγt mRNA相对表达水平为(7.96±3.09)×10-5,明显高于对照组[(4.19±2.07)×10-5],外周血Foxp3 mRNA相对表达水平为(3.79±1.25)×10-4,明显低于对照组[(15.89±7.51)×10-4],差异均有统计学意义(P<0.05).研究组患儿血清TGF-β 水平为(58.58±12.83)ng/L,明显低于对照组[(8.81±3.97)ng/L],IL-17 水平为(22.09±10.28)ng/L,明显高于对照组[(80.14±8.95)ng/L],差异均有统计学意义(P<0.05).结论 EBV感染相关病症发生及发展中Th17、Treg细胞发挥着重要作用,增大Th17、Treg细胞及其相关炎症因子监测力度可为EBV感染患儿的疾病情况及疗效评估提供参考依据.
Objective To study the clinical significance of Th17 and Treg cell detection in children with Epstein-Barr virus(EBV)in-fection.Methods By retrospective analysis,60 children with EBV infection from January 2021 to January 2023 in Luzhou People's Hospital were selected as the study group,and 60 healthy children admitted for physical examination during the same period were selected as the control group.Th17 and Treg cells in peripheral blood T lymphocytes were determined by flow cytometry,and the levels of related characteristic cytokines[trans-forming growth factor-β(TGF-β)and interleukin-17(IL-17)]were determined by enzyme-linked immunosorbent antibody double sand-wich method.Meanwhile,the mRNA expression levels of Th17 expressing solitary nucleus receptor(RORγt)mRNA and Treg expressing forks wing spiral transcription factor(Foxp3)was determined by RT-PCR.The expression rates of Treg cells,Th17 cells,TGF-β,IL-17 and RORγt mRNA and Foxp3 mRNA were compared between the two groups.Results The expression rate of peripheral blood Th17 cells in the study group was(1.39±0.42)%,which was significantly higher than that in the control group[(0.82±0.19)%],and the expression rate of periph-eral blood Treg cells was(3.48±0.84)%,which was significantly lower than that in the control group[(6.07±1.25)%],the differences were statistically significant(P<0.05).The relative expression level of peripheral blood RORγt mRNA in the study group was(7.96±3.09)x 10-5,which was significantly higher than that in the control group[(4.19±2.07)× 10-5],the relative expression level of Foxp3 mRNA in pe-ripheral blood was(3.79±1.25)x10-4,which was significantly lower than that in the control group[(15.89±7.51)× 10-4],the differ-ences were statistically significant(P<0.05).The level of serum TGF-β in the study group was(58.58±12.83)ng/L,which was signifi-cantly lower than that in the control group[(8.81±3.97)ng/L],and the level of IL-17 was(22.09±10.28)ng/L,which was significantly higher than that in the control group[(80.14±8.95)ng/L],the differences were statistically significant(P<0.05).Conclusion Th 17 and Treg cells play an important role in the occurrence and development of EBV infection-related diseases.Increasing the monitoring of Th17 and Treg cells and their associated inflammatory factors can provide reference for the disease status and efficacy evaluation of children with EBV infection.
