目的 探究肝损伤介导肝星形细胞小窝蛋白-1的表达的影响.方法 通过肝总管结扎手术诱导大鼠肝损伤模型和体外培养肝星形细胞LX-2,检测肝组织和肝星形细胞小窝蛋白-1的表达水平.结果 ①与假手术组相比,胆总管结扎手术可显著诱导肝脏损伤,手术后7d,手术组大鼠体重显著减轻,肝脏重量和肝指数显著增加,以及血清谷丙转氨酶和谷草转氨酶水平显著增加;②与假手术组相比,肝损伤可显著诱导小窝蛋白-1在mRNA和蛋白水平显著增加;③与胆总管结扎组相比,LPS处理可显著增加肝星形细胞小窝蛋白-1的表达水平.结果 肝损伤可显著诱导肝星形细胞小窝蛋白-1的表达.
Objective This study was to investigate the effect of liver injury-mediated caveolin-1 expression in hepatic stellate cells (HSCs).Methods Through establishing bile duct ligation (BDL)-induced liver injury model in vivo and culturing HSCs line LX-2 in vitro,and then we measured the caveolin-1 expression in liver injury tissue and LX-2.Results The results showed:1) compared with sham surgery group,BDL can remarkably induce liver injury in rat evidenced by significant decreased body weight,increased liver weight and liver index,and increased serum alanine transaminase (ALT) and aspartate aminotransferase (AST) levels in rats;2) compared with sham surgery group,liver injury can significantly induce caveolin-1 expression both in mRNA and protein levels;3) compared with unstimulated group,LPS treated group showed higher caveolin-1 expression levels in LX-2.Conclusion Our study suggest that liver injury can observably induce caveolin-1 expression in HSCs.
