目的 探讨干扰素-γ(IFN-γ)基因+874位点单核苷酸基因多态性与IgA肾病(IgAN)患者临床检验指标间的关系.方法 从131例IgAN患者和138例健康对照者外周血中提取DNA,采用序列特异性引物聚合酶链反应(PCR-SSP)技术检测IFN-γ基因+874位点单核苷酸基因多态性,比较两组的基因型和等位基因的分布,分析基因型与IgAN临床检验数据的关系.结果 IgAN患者不同基因型组间24h尿蛋白定量差异有统计学意义(P<0.05).结论 IFN-γ基因+874位点单核苷酸多态性与24h尿蛋白定量存在相关性.
Objective To investigate the relationship between interferon-γ(IFN-γ) gene+874 locus single nucleotide polymorphisms and clinical test in IgA nephropathy(IgAN) patients.Methods DNA was extracted from peripheral blood from 131 IgAN patients and 138 healthy controls.Sequence specific primer polymerase chain reaction (PCR-SSP) was used to detect the single nucleotide polymorphisms in the +874 locus of IFN-γgene.The distribution of genotypes and alleles in the two groups was compared.And the relationship between the genotype and the clinical data of the IgAN was analyzed.Results There was a significant difference in 24 h urine protein in IgAN patients with different genotypes (P<0.05).Conclusions There was a correlation between the single nucleotide polymorphisms at the +874 site of IFN-gamma and the quantitative 24 h urine protein.
