不同教材对"DNA的粗提取与鉴定"实验步骤的表述存在差异,留下了疑问.本研究进一步分析了实验原理,并进行了多组平行实验,探索实验步骤的优化.结果表明,比较合适的研磨液NaCl浓度为2mol/L,三种去杂质方式对实验结果影响不大,不溶解DNA沉淀物而直接用二苯胺鉴定的效果更明显.
Different textbooks have different descriptions of experimental steps of"Coarse Extraction and Identification of DNA,"which raises questions.This study further analyzed the experimental principles and conducted multiple parallel experiments to explore optimization of the experimental steps.The results showed that a suitable concentration of NaCl in the grinding solution is 2 mol/L,three impurity removal methods have little effect on the experimental results,and the effect of direct identification using diphenylamine without dissolving DNA precipitate is more significant.
