[目的]研究苯丙噻二唑(BTH)处理对马铃薯愈伤早期活性氧产生的影响.[方法]用100 mg/L BTH处理模拟损伤的大西洋马铃薯块茎,测定BTH处理后不同时间块茎伤口处胞内钙离子浓度、活性氧产生关键酶的基因表达量和活性及超氧阴离子和过氧化氢含量.[结果]BTH处理增加了愈伤早期马铃薯块茎伤口处组织的胞内Ca2+浓度,促进了StCDPK14的表达,提高了CDPK活性.BTH显著上调了Strbohs家族成员的表达,StrbohA在4 h时达到峰值,StrbohB、StrbohC、StrbohD和StrbohH在8 h时达到峰值.BTH还显著上调了StSODs家族成员的表达,StCSD1表达量随着处理时间的延长而升高,其余成员在8 h时达到峰值.由于Strbohs和StSODs的上调表达,明显促进了块茎伤口处O2·-和H2O2的产生.[结论]BTH可通过提高胞内钙浓度来激活CDPK,CDPK进一步激活Strbohs和StSODs促进活性氧的产生.
[Objective]The study aimed to explore the effect of benzo(1,2,3)-thiadiazole-7-carbo-thioic acid(BTH)treatment on reactive oxygen species(ROS)production at wounds of potato tubers dur-ing early stage of healing.[Method]The wounded potato tubers(cv.Atlantic)were treated with 100 mg/L BTH,followed by determining intracellular calcium ion concentration,gene expression and activities of key enzymes for reactive oxygen species production,and superoxide anion and hydrogen peroxide content in the tuber wounds at different time after BTH treatment.[Result]The concentration of intracellular Ca2+ in-creased after BTH treatment.Meanwhile,BTH promoted the expression of StCDPK14 and enhanced the calcium-dependent protein kinases(CDPK)activity in the wounds of potato tubers.BTH significantly up-regulated the expression of Strbohs family members,StrbohA peaked in 4 h after treatment,and StrbohB,StrbohC,StrbohD and StrbohH peaked in 8 h.BTH also significantly upregulated the expression of St-SODs family members,and the expression of StCSD1 increasing with the extension of BTH treatment time,and the rest members peaked in 8 h after treatment.The upregulated expression of Strbohs and StSODs led to a remarkable increase in the contents of O2·-and H2O2 in the wounds.[Conclusion]BTH treatment could activate CDPK activity by increasing intracellular calcium concentration,which further upregulated Strbohs and StSODs expressions,promoting reactive oxygen species production at early stage of healing.
