目的 探讨人脂肪干细胞(adipose-derived stem cells,ADSCs)免疫共抑制分子PD-L1和Gal-9的表达及其影响因素.方法 培养并鉴定28例行吸脂术健康女性ADSCs,采用流式细胞术检测PD-L1和Gal-9表达,采用多因素线性回归分析年龄、身高体重指数(BMI)、脂肪采集部位以及干扰素-γ(interferon-γ,IFN-γ)对ADSCs中PD-L1和Gal-9表达水平的影响.结果 体外培养细胞符合ADSCs国际标准,且表达PD-L1和Gal-9.17例乳腺ADSCs中PD-L1和Gal-9的表达分别为37.24%±8.20%和4.41% ±2.65%,显著高于11例腹部的28.80%±8.59%和2.51%±1.39%(P=0.018,P=0.039).多因素线性回归分析显示,年龄和采集部位是影响PD-L1表达的独立危险因素(P=0.009,P=0.006),年龄每增加1岁PD-L1表达降低0.385%,乳腺部位比腹部高8.58%.采集部位是影响Gal-9表达的独立危险因素(P=0.041),乳腺部位比腹部高1.898%.BMI不是ADSCs表达PD-L1和Gal-9的影响因素.经20 ng/ml IFN-γ体外刺激48 h后,ADSCs的PD-L1阳性细胞百分比为49.850%±4.414%,显著高于刺激前(30.790%±3.602%,P=0.0036);Gal-9阳性细胞百分比为5.084%±0.619%,显著高于刺激前(2.173%±0.267%,P=0.0004).结论 人ADSCs表达PD-L1和Gal-9,且供者年龄小、采集乳腺部位及IFN-γ显著增加PD-L1和Gal-9表达.
Objective To explore whether human adipose-derived stem cells (ADSCs) express PD-L1 and Gal-9 and its potential influence factors.Methods ADSCs isolated from 28 healthy female donors who underwent liposuction of the abdomen or breast tissue were cultured and characterized.The expression of PD-L1 and Gal-9 were detected using flow cytometry.The impact of donor age,body mass index (BMI),donor site and interferon-γ (IFN-γ) on the expression of PD-L1 and Gal-9 was analyzed using multivariate linear regression analysis.Results The cultured cells fulfilled the criteria for defining MSCs according to the international standards and expressed PD-L1 and Gal-9.Breast-derived ADSCs had higher expression levels of PD-L1 (37.24% ± 8.20%) and Gal-9 (4.41% ± 2.65%) than those in abdomen-derived ADSCs (28.80% ± 8.59% and 2.51% ± 1.39%,respectively) (P =0.018,P =0.039).Multivariate linear regression analysis showed that donor age and site were independent risk factors affecting PD-L1 expression (P =0.009,P =0.006).The expression of PD-L1 decreased by 0.385% for every one year of age increase,and it was 8.58% higher in the breast-derived ADSCs than in the abdomen-derived ADSCs.Donor site was an independent risk factor for Gal-9 expression (P =0.041).Gal-9 positive expression rate in the breast-derived ADSCs was 1.898% higher than that in the abdomen-derived ADSCs.BMI was not a risk factor affecting PD-L1 or Gal-9 expression on ADSCs.The expression of PD-L1 and Gal-9 on ADSCs were significantly upregulated after 48 hours stimulation with 20 ng/ml IFN-γin vitro.Conclusions Human ADSCs expressed PD-L1 and Gal-9.Donor age,site and IFN-γ treatment were independent risk factors affecting the expression of PD-L1 and Gal-9 on ADSCs.
