目的 通过局部注射不同剂量的骨髓基质细胞(bone marrow stromal cells,BMSCs)条件培养液神经活性蛋白,观察其对大鼠坐骨神经损伤后的再生作用.方法 取Wistar大鼠30只,随机分成3组,每组10只.行左侧坐骨神经切断并造成6mm缺损,用10 mm硅胶管套接缝合.其中10只硅胶管内注射高剂量BMSCs条件培养液神经活性蛋白(高剂量组),10只硅胶管内注射低剂量BMSCs条件培养液神经活性蛋白(低剂量组),10只硅胶管内注射等量生理盐水(对照组).术后8周通过后肢小腿三头肌肌湿重、形态学及图像分析为观察指标来判断不同剂量的BMSCs条件培养液神经活性蛋白对大鼠坐骨神经损伤后再生作用的影响.结果 高剂量组和低剂量组各项指标均优于对照组,高剂量组各项指标均优于低剂量组.结论 BMSCs条件培养液神经活性蛋白对坐骨神经再生有明显作用,高剂量作用更佳.
Objective To observe the effect of local injection of different dosages of neurotrophi(e) proteins from bone marrow stromal cells (BMSCs) conditioned medium on nerve regeneration after sciatic nerve injury in rats. Methods Thirty Wister rats were used in the study where the left sciatic nerve was resected to create a 6 mm gap which was bridged with a 10 mm-long silicon tube.The rats were randomly divided into three groups with 10 each.High dosage of neurotrophic proteins from BMSCs conditioned medium was injected in the silicon tube of the high dosage group,while low dosage of neurotrophic proteins was injected in the silicon tube of the low dosage group.0.9% normal saline was injected in the silicon tube of the control group.After 8 weeks,tricepital muscle wet weight and morphology and neuromorphometry analysis were conducted to assess the effect on sciatic nerve regeneration by different doses of neurotrophic proteins from the conditioned medium of BMSCs.Results The indices from the high dosage group and low dosage group were superior to those from the control group.The indices from the high dosage group were superior to those from the low dosage group. Conclusion Neurotrophic proteins from the conditioned medium of BMSCs have obvious effects in promoting sciatic nerve regeneration.The higher the dosage,the better the effect.
