建立葛根素的高效液相色谱检测方法.使用70%乙醇溶液微波辅助提取,C18色谱柱分离,二极管阵列检测.利用Box-Behnken响应面分析优选微波提取时间、温度和溶剂组成,得到最佳提取工艺为:微波提取时间9.7 min、提取温度60.7 ℃、乙醇-水比例为71.2%,在此条件下,实验优化后葛根素平均提取率达2.09 %.葛根素在1.0~200 mg/L范围内呈现良好的线性关系,相关线性系数r为0.9997,检出限为0.3 mg/L,加标回收率为97.3%~99.2%,相对平均偏差RSD为3.72%~6.21%.
A method was established for determination of puerarin in gegen. 70% alcohol was used in microwave assisted extraction process, C18 chromatographic column was used for separation and the analytes were detected by diode array detection. The process parameters including extraction time, extraction temperature and solvent constitution were optimized by Box-Behnken response surface methodology (RSM) analysis experiment. The results of response surface methodology showed that the optimal operation conditions were as follows: extraction time 9.7 min, extraction temperature 60.7 ℃, the ratio of ethanol to water 71.2%. The final extraction rate of this method was 2.09%. There was a good linear relationship in the certain range of 1.0~200 mg/L(linear regression coefficient: 0.999 7) and the limit of detection was 0.3 mg/L. The recoveries were in the range of 97.3%~99.2%,and RSDs were 3.72%~6.21%.
