目的 探讨阿司匹林对胃癌干细胞有无抑制作用及可能机制.方法 用RMPI-1640培养液培养人胃癌细胞株SGC7901,无血清培养基悬浮培养抽提富集微球体.MTT法检测阿司匹林对SGC7901细胞微球体生长的抑制情况.平板克隆实验观察阿司匹林对SGC7901微球体细胞克隆形成的影响.Transwell小室实验观察阿司匹林对SGC7901微球体迁移侵袭能力的影响.Western blot检测经过阿司匹林孵育后胃癌干细胞E2F1和Sox2蛋白的表达情况.结果 阿司匹林对人胃癌细胞株SGC7901微球体细胞的增殖、克隆形成、迁移和侵袭能力具有显著的抑制作用.阿司匹林可剂量依赖性下调胃癌干细胞中E2F1和Sox2蛋白的表达.结论 阿司匹林对人胃癌细胞株SGC7901微球体细胞的恶性行为具有抑制作用,其抑制作用与E2F1和Sox2蛋白表达下降有关.
Objective To investigate the inhibitory effect of aspirin on gastric cancer stem cells and its possible mecha-nism.Methods Human gastric cancer cell line SGC7901 was cultured in RMPI-1640 medium,and microspheres were extracted and enriched by suspension culture in serum-free medium.The MTT method was used to detect the inhibition of aspirin on the growth of SGC7901 cell microspheres.The plate cloning experiment was performed to observe the effect of aspirin on the colony for-mation of SGC7901 microsphere cells.Transwell chamber experiment was performed to observe the effect of aspirin on the migration and invasion ability of SGC7901 microspheres.Expression of proteins E2F1 and Sox2 in gastric cancer stem cells after incubation with aspirin was detected by Western blot.Results Aspirin had a significant inhibitory effect on the proliferation,colony formation,migration and invasion of human gastric cancer cell line SGC7901 microsphere cells.Aspirin can dose-dependently down-regulate the expression of E2F1 and Sox2 proteins in gastric cancer stem cells.Conclusion Aspirin has an inhibitory effect on the migration and invasion of human gastric cancer cell line SGC7901,and its inhibitory effect is related to the decrease in the expression of E2F1 and Sox2 proteins.
