为确定鸡TLR1A基因g.926G>A和g.1076G>C等2个nsSNP位点多态性及其功能意义,采用CRS-PCR-RFLP方法检测了 310只坝上长尾鸡(阳性127只,阴性183只)TLR1A基因2个nsSNP位点的多态性、二者与沙门氏菌抗性的关联,并对其进行生物信息学与适应性进化分析.结果表明,g.926G>A(S309N)和g.1076G>C(R359P)与鸡沙门氏菌易感性显著相关(P<0.05).其中,S309N在种间种内水平均受到正选择压力;R359P在种内受到正选择压力.二者均具有有害性,且后者蛋白稳定性较低.鸡TLR1A基因的g.926G>A和g.1076G>C位点可作为鸡抗病育种中潜在的功能性位点.
The polymorphism of two nsSNP loci of TLR1A gene in 310 Bashang long-tailed chickens(127 pos-itive and 183 negative),the association between the two and Salmonella resistance,and the bioinformatics and adaptive evolution were studied with CRS-PCR-RFLP method to explore the polymorphism and functional sig-nificance of two nsSNPs of g.926g>a and g.1076g>c in chicken TLR1A gene.The results showed that g.926G>A(S309N)and g.1076G>C(R359P)were significantly correlated with the susceptibility of chicken Salmonella(P<0.05).S309N was under positive selection pressure in interspecific and intraspecific levels;R359P was under positive selection pressure within species.Both of them were harmful,and the protein sta-bility of the latter was low.The g.926g>a and g.1076g>c loci of chicken TLR1A gene could be used as po-tential functional loci in chicken disease resistance breeding.
