目的 探讨补肾活血方对佐剂性关节炎(Adjuvant arthritis,AA)大鼠血清中细胞因子基质金属蛋白酶-3(Matrix Metalloproteinase 3,MMP-3)与白细胞介素-4(Interleukin-4,IL-4)、白细胞介素-10(Interleukin-10,IL-10)的影响.方法 以雷公藤多苷片为阳性对照药物,给予AA大鼠不同剂量的补肾活血方制成药液灌胃,治疗28 d,观察治疗前后各组大鼠的一般情况、体重变化、关节炎指数,并测定各组大鼠血清中MMP-3、IL-4、IL-10水平.结果 与空白对照组比较,模型对照组大鼠体重增长缓慢,血清中MMP-3显著升高,而IL-4及IL-10则明显下降,差异有统计学意义(P<0.01);与模型对照组比较,给药后的中药不同剂量组及雷公藤多甙片组(对照组)大鼠体重增长较快,关节炎指数降低明显,且均可降低MMP-3,升高IL-4、IL-10水平,差异有统计学意义(P<0.01);与对照组比较,中药高剂量组能显著增加AA大鼠体重(P<0.05),升高IL-4、IL-10水平(P<0.05),降低关节炎指数及血清中MMP-3的水平(P<0.05).结论 补肾活血方通过增加IL-4、IL-10水平,降低血清中MMP-3含量,恢复类风湿性关节炎细胞因子间的平衡,从而达到防治该病的作用.
Objective To observe the impacts of bushen huoxue formula on MMP-3,IL-4 and IL- 10 in serum of the rats of adjuvant arthritis. Methods Tripterygium glycosides tablets were taken as the positive reference medicine. Bushen huoxue formula of different doses was prepared as liquid for lavage in the rats of adjuvant arthritis. After treatment for 28 days,the general situation,body weight and arthritis index were observed before and after treatment. The levels of MMP-3,IL-4 and IL-10 were determined in each group. Results Compared with the blank control group,the body weights were increased slowly,MMP-3 in-creased significantly and IL-4 and IL-10 decreased remarkably in the model control group,indicating the significant differences(P < 0. 01). Compared with the model control group,in the Chinse herbal medicine groups of different doses and tripterygium glycosides tablets group(control group),the body weights were in-creased quickly,the arthritis indexes decreased remarkably,MMP-3 decreased and IL-4 and IL-10 in-creased,indicating the significant differences(P < 0. 01). Compared with the control group,the Chinese herb-al medicine group of high dose,the body weights of rats were increased significantly(P < 0. 05),the levels of IL-4 and IL-10 increased(P < 0. 05),the arthritis indexes and MMP-3 decreased(P < 0. 05). Conclu-sion Bushen Huoxue formula recovers the balance among the cytokines in rheumatoid arthritis through in-creasing IL-4 and IL-10 and reducing MMP-3 in serum so as to prevent and treatment the disease.
