目的 研究黄甲软肝颗粒对小鼠醉酒的解酒作用和CCl4 诱导小鼠急性肝损伤的护肝作用.方法 取SPF级雄性昆明小鼠60 只,按体质量随机分为正常组,模型组,低、中、高剂量黄甲软肝颗粒组,谷胱甘肽组,每组10 只.除正常组与模型组灌服相同体积的蒸馏水外,其余各组给予相对应剂量的药物,正常组和模型组给予 20 mL/kg蒸馏水,1 次/d,连续7d.末次给药前各组小鼠禁食12 h,末次给药60 min后,除正常组外的各组按14 mL/kg给予60°红星二锅头白酒灌胃.60 min后,取小鼠血液及肝脏组织,测定血中乙醇浓度,测定肝脏组织中谷胱甘肽、MDA含量及ADH活力,观察黄甲软肝颗粒解酒作用.另取SPF级雄性昆明小鼠 60 只,按体质量随机分为正常组,模型组,低、中、高剂量黄甲软肝颗粒组,阳性对照药谷胱甘肽组,每组 10 只,连续给药 14 d,动物末次给药后禁食 16 h,模型组及各给药组灌胃 0.5%CCl4 植物油溶液5 mL/kg,正常组灌胃植物油5 mL/kg.24 h后,摘眼球取血及分离保存肝脏组织用于指标检测和组织病理学检查.观察黄甲软肝颗粒护肝作用.结果 与模型组比较,黄甲软肝颗粒高剂量组和谷胱甘肽组小鼠血液中乙醇浓度显著降低(P<0.05).与模型组比较,黄甲软肝颗粒高剂量组谷胱甘肽组的谷胱甘肽含量显著增加(P<0.05);黄甲软肝颗粒中剂量组和谷胱甘肽组MDA含量显著降低(P<0.05);黄甲软肝颗粒高剂量组和谷胱甘肽组ADH活性显著增加(P<0.05).与模型组比较,黄甲软肝颗粒中剂量组和谷胱甘肽组血清ALT、AST活性均显著降低(P<0.05 或P<0.01);黄甲软肝颗粒中剂量组和谷胱甘肽组肝脏组织MDA含量显著降低(P<0.05).谷胱甘肽组的谷胱甘肽含量显著增加(P<0.05).与模型组比较,黄甲软肝颗粒中、高剂量组和谷胱甘肽组小鼠肝细胞结构有一定改善,细胞浆和细胞核溶解程度有所减轻.结论 黄甲软肝颗粒能降低急性酒精性肝损伤小鼠血液中乙醇浓度水平,减轻肝组织氧化损伤;能降低CCl4 急性肝损伤小鼠的肝功能损伤和氧化损伤,改善肝组织细胞病变,黄甲软肝颗粒具有较好的解酒护肝作用.
Objective To study the antialcoholic effect of Huangjia Ruangan Granules on mice in-toxication and the hepatoprotective effect of CCl4-induced acute liver injury in mice.Methods 60 SPF grade male Kunming mice were randomly divided into normal control group,model control group,low,medium and high dose Huangjia Ruangan Granules group and positive control gluta-thione group,with 10 mice in each group.Except for the normal control group and the model control group,which were infused with the same volume of distilled water,the remaining groups were given the corresponding doses of drugs.The normal group and model group were given 20mL/kg distilled water once a day for 7 consecutive days.Before the last administration,the mice in each group were fasted for 12 h,and after the last administration for 60 min,all groups except the normal group were given 60°red star Erguotou white wine at the rate of 14 mL/kg.After 60 min,the blood and liver tissues of mice were taken to determine the concentra-tion of alcohol in blood,the content of glutathione and MDA in liver tissues and the activity of ADH,and the antialcohol effect of Huangjia Ruangan Granules was observed.60 SPF grade male Kunming mice were randomly divided into normal control group,model control group,low,me-dium and high dose Huangjia Ruagan Granules group and positive control glutathione group with 10 mice in each group according to body weight.The drug was administered continuously for 14 d.After the last administration,fasting was performed for 16h.The model group and the drug ad-ministration groups were gavaged with 0.5%CCl4 vegetable oil solution,5 mL/kg,and the nor-mal group was gavaged with 5 mL/kg of vegetable oil.24 h later,blood was collected from the eyeballs and liver tissue was separated and preserved for the index test and histopathological examination.The protective effect of Huangjia Ruangan Granules on liver was observed.Results Compared with alcohol acute liver injury model group,the concentration of alcohol in blood of mice in high dose Huangjia Ruangan Granules group and glutathione group was significantly de-creased(P<0.05).Compared with model group,glutathione content in glutathione group was significantly increased(P<0.05).MDA content was significantly decreased in medium dose group and glutathione group(P<0.05).ADH activity was significantly increased in high dose groups and glutathione groups(P<0.05).Compared with model group,ALT activity in serum of medium dose group was significantly decreased(P<0.05).AST activity was significantly de-creased(P<0.05).Serum ALT and AST activities in glutathione group were significantly de-creased(P<0.05 or P<0.01).The content of MDA in liver tissue of medium dose group and glutathione group was significantly decreased(P<0.05).The glutathione content in glutathione group was significantly increased(P<0.05).Compared with the model group,the liver cell structure of mice in medium and high dose groups and glutathione group was improved to some extent,and the degree of cytoplasmic and nuclear dissolution was reduced.Conclusion Huangjia Ruangan Granules can reduce the concentration of alcohol in blood and the oxidative damage of liver tissue in mice with acute alcoholic liver injury.It can reduce liver function dam-age and oxidative damage in mice with CCl4 acute liver injury and improve liver tissue cytopath-ia.Huangjia Ruangan Granules has a good effect on relieving alcohol and protecting liver.
