细胞质雄性不育(CMS)在薹用白菜育种中己被广泛使用,为提高其CMS类型鉴定的准确性和效率,探究当前薹用白菜育种中CMS类型的应用特征,首先针对己报道在薹用白菜中应用的Polima,Ogura与Hau类型的CMS开发了特异性多重PCR分子标记Brcms_M.该标记解决了现有Polima分子标记在Hau CMS材料中的假阳性问题,可同时将3种CMS类型与正常胞质类型区分开.利用Brcms_M对28个薹用白菜细胞质雄性不育杂交品种/组合进行鉴定,结果表明Polima是当前红菜薹育种中主要应用的CMS类型,白菜薹与菜心CMS则均以Ogura类型为主,同时发现2个存在不完全败育现象且不含恢复基因Rfo的Ogura CMS品种.
Cytoplasmic male sterility(CMS)has been widely applied in flowering Chinese cabbage breeding.To improve the accuracy and efficiency of CMS type identification and investigate the CMS application in flowering Chinese cab-bage,the specific multiplex PCR CMS marker Brcms_M for simultaneously detecting Polima,Ogura and Hau CMS was developed.Brcms_M could overcome the false positive problem in Polima CMS identification when using the reported Polima markers in Hau CMS materials,and also could distinguish three CMS and normal cytoplasmic type.Brcms M was furtherly used to identify the CMS type of the twenty-eight flowering Chinese cabbage varieties.The result of CMS type identification indicated that Polima CMS was the mainly applied in purple flowering Chinese cabbage breeding,and CMS type of white flowering Chinese cabbage and flowering Chinese cabbage majorly belong to the Ogura CMS.Inter-estingly,two partial sterility varieties were also identified as Ogura CMS type,Ogura CMS restorer gene Rfo was not found in these two varieties.
