目的 研究基因层面信息与胃癌淋巴结转移的相关性及 14-3-3β蛋白是否参与胃癌淋巴转移.方法 收集2022 年1~6 月间30 例在哈尔滨医科大学附属肿瘤医院胃肠外科行胃癌根治术患者的胃癌组织及癌旁组织,依据术后病理结果分为淋巴结转移阳性组(Pos组)和淋巴结转移阴性组(Neg组)2 组,每组各15 例.利用转录组测序技术获得胃癌患者样本的转录本序列信息,使用生物信息学分析方法比较胃癌患者在有无淋巴转移时的差异表达基因,并进一步分析其生物学意义.结果 ①Pos组和Neg组间共1 878 个基因表达水平有显著差异,其中差异高表达基因 683 个,差异低表达基因 1 195个,差异显著性最明显的两个基因是USP24P1(P =7.6760e-03)和DEFA5(P =1.8585e-02);②基因本体(Gene Ontology,GO)富集分析显示,差异基因主要富集在突触传递的调节、突触间信号传递、肌肉系统、含胶原蛋白的细胞外基质、突触膜、突触后膜、阳离子通道、丝氨酸型肽活性、丝氨酸水解酶活性等功能条目上;③Pos组活化的CD4+T细胞显著高于Neg组(P =0.0329),而未成熟的树突状细胞显著低于Neg组(P =0.0235);④YWHAB基因表达的14-3-3β蛋白在两组间数据统计分析显示,两组间未发现显著差异(P =0.68).结论 基因USP24P1、DEFA5 及活化的CD4+T细胞与胃癌的淋巴结转移存在相关性.
Objective To study the correlation between gene level information and lymph node metastasis of gastric cancer and whether 14-3-3-β protein is involved in lymph node metastasis of gastric cancer.Methods The gastric cancer tissues and adjacent tissues of 30 patients un-dergoing radical gastrectomy in the Department of Gastrointestinal Surgery of the Harbin Medi-cal University Cancer Hospital from January 2022 to June 2022 were collected.According to postoperative pathological results,the patients were divided into two groups:positive lymph node metastasis group(Pos group)and negative lymph node metastasis group(Neg group),with 15 cases in each group.Bioinformatics analysis was used to comprehensively and rapidly obtain almost all the transcriptome sequence information of the samples of gastric cancer pa-tients.The differential expression genes of gastric cancer patients with and without lymphatic metastasis were compared by statistical methods.Its biological significance was further ana-lyzed.Results ①There were significant differences in the expression levels of 1 878 genes between Pos and Neg groups,including 683 genes with high expression and 1 195 genes with low expression.The two genes with the most significant differences were USP24P1(P = 7.6760e-03)and DEFA5(P = 1.8585e-02);②Gene Ontology(GO)enrichment analysis showed that the differential genes were mainly enriched in the regulation of synaptic transmis-sion,inter-synaptic signal transmission,muscle system,extracellular matrix containing colla-gen,synaptic membrane,postsynaptic membrane,cation channel,serine type peptide activity,serine hydrolase activity and other GO terms;③Activated CD4+T cells in the Pos group were significantly higher than those in the Neg group(P =0.0329),while immature dendtric cells were significantly lower than those in the Neg group(P =0.0235);④Statistical analysis of 14-3-3-β protein expression of YWHAB gene between the two groups showed that P =0.68,and no significant difference was found between the two groups.Conclusion Genes USP24P1,DEFA5 and activated CD4+T cells are associated with lymph node metastasis of gastric cancer.
