1. ÖZET Bu çalışmada, naftalinin farelerde meydana getirdiği oksidan hasara karşı sarımsak (Bulbus Allii sativi) ekstresinin antioksidan etkisi bazı biyokimyasal parametreler ve histopatolojik incelemelerle değerlendirildi. Çalışmamızda 25-30 g ağırlığında 50 adet Balb-c türü dişi/erkek fareler kullanıldı. Fareler; 1. grup: Kontrol (K), 2. grup: Mısır yağı (MY), 3. grup: Sarımsak (S), 4. grup: Naftalin (N), 5. grup Naftalin + Sarımsak (N+S), olmak üzere 5 gruba ayrıldı. Bir ay süreyle (6 gün/hafta) standart yem ile normal diyete tabi tutulan gruplardan, K grubu hiçbir işlem görmedi, MY grubuna 3.6 g/kg dozda mısır yağı, S grubuna 250 mg/kg dozda sarımsak ekstresi, N grubuna 100 mg/kg dozda mısır yağında çözündürülmüş naftalin, ve N+S grubuna 125 mg/kg dozda sarımsak ekstresi + 100 mg/kg dozda naftalin intraperitonal uygulandı. Bir ay sonunda dekapite edilen hayvanların karaciğer ve böbreklerinden doku örnekleri alındı. Bu örneklerde oksidan hasan değerlendirmek amacı ile lipid peroksidasyonun son ürünü olan malondialdehit (MDA) ve antioksidan glutatyon (GSH) tayinleri ve ayrıca histopatolojik incelemeler yapıldı. K, MY ve S gruplarında karaciğer ve böbrek dokularında ortalama (ort.) GSH ve MDA değerleri birbirlerine yakın bulundu. N grubunda ise ort. GSH değerleri kontrol grubuna göre anlamlı bir şekilde düşerken, bu gruba ait ort. MDA değerleri ise anlamlı bir şekilde artış gösterdi. N+S grubunun karaciğer ve böbrek dokularının ort. GSH değerleri N grubuna göre anlamlı olarak artarken ort. MDA değerleri İse anlamlı derecede azalma gösterdi. Bu bulgular sonucunda naftaline bir ay süreyle maruziyetin karaciğer ve böbrekte oksidan doku hasarına neden olduğu, sarımsak {Bulbus Allii sativi) ekstresinin antioksidan etkisi ile oksidan hasan büyük ölçüde azalttığı düşünülmektedir. Histopatolojik incelemeler de biyokimyasal verileri desteklemektedir. 2. SUMMARY INVESTIGATION OF NAPHTHALENE - INDUCED OXIDATIVE STRESS AND ANTIOXD3ATIVE EFFECT OF GARLIC (Bulbus Allii sativi) EXTRACT The aim of this study was to investigate the protective effects of garlic {Bulbus Allii sativi) extract against naphthalene-induced oxidative stress using biochemical and histological analysis. For this purpose 50 Balb-c male/female mice each weighing 25-30 g were used. Animals were divided into 5 groups as follows; Control group (C): Mice were only fed with standard laboratory chow; Corn oil group (CO): CO (3.6 g/kg) was administered intraperitoneally for 30 days (6 days/week); Aqueous garlic extract group (AGE): 250 mg/kg AGE was administered intraperitoneally for 30 days (6 days/week); Naphthalene group (N): N in corn oil (100 mg/kg) was administered intraperitoneally for 30 days (6 days/week); Naphthalene + AGE group (NAGE): N in corn oil (100 mg/kg) + AGE extract (125 mg/kg) was administered intraperitoneally for 30 days (6 days/week). After 30 days, mice were decapitated and hepatic and renal tissue samples were obtained for the histopathological analysis of damage and the measurement of malondialdehyde (MDA) - indicating lipid peroxidation- and glutathione (GSH) -a key antioxidant- levels For C, CO and AGE groups, mean glutathione (GSH) levels and malondialdehyde (MDA) levels of hepatic and renal tissue was similar to each other. Mean GSH levels of N group was significantly lower and mean MDA levels of N group was significantly higher than those of the C group. However, AGE administration to the naphthalene group reversed these effects significantly and the GSH levels of N + AGE group was found significantly higher and mean MDA levels of this group was found significantly lower than those of the N group which were close to C group. In the light of these findings, we can suppose that naphthalene induces oxidative stress in the hepatic and renal tissues of Balb-c mice and garlic (Bulbus Allii sativi) extract is significantly decreased this oxidative stress by its antioxidant effects. Our biochemical data are also supported by histological investigations. 70
