Bu çalışmada, çevresel kirliliğin önemli etmenlerinden BAP'ın testiste oluşturabileceği hasarda antioksidan olarak yeğlenen Curcumin'in olası koruyucu etkilerinin immünohistokimyasal yöntemlerle incelenmesi amaçlandı.Araştırmamızda, 36 adet Wistar albino erkek sıçan,her grupta 6 denek olacak şekilde 6 gruba ayrıldı 1. Grup: Kontrol, 2. Grup: Mısır yağı, 3. Grup: DMSO, 4. Grup: BaP, 5. Grup: Curcumin, 6. Grup: BaP +Curcumin olarak belirlendi. 6 hafta sonunda deneklerin testis dokuları alınarak alışılmış ışık mikroskobik izleme yöntemlerinden geçirildi. Kesitlerin bir kısmı Hematoksilen-Eozin, bir kısmı Leydig hücrelerindeki değişiklikleri izleyebilmek için AT1 ve AR dağılımınıincelemek için AR primer antikorlarıyla indirekt immünohistokimyasal yöntemle boyandı. Spermatogenezis ve spermiyogenezis sürecinde seminifer epitel ve özellikle Leydig hücreleri için önemli bir işaretleyici olan AT1 immünreaksiyonunun BaP uygulaması ile arttığı, Curcumin uygulaması ile kısmen geri döndüğü saptandı. Bu bulgunun ışığında BaP'ın testosteron sentezini olumsuz etkilemiş olabileceği düşünüldü. Erkek üreme sisteminde önemli etkileri olan AR immünreaktivitesininçekirdekteki tutulumu, BaP gruplarında genellikle adluminal bölgede bulunan hücrelerdeydi. Bazı birleşmiş spermatidlerde orta dereceli immünreaktivite izlendi. BaP+Curcumin grubunda, BaP'a karşın tepkimenin kontrollere yaklaştığı izlendi. Bu bulgu BaP'ın spermiyogenezisi olumsuz etkilemiş olabileceğini düşündürdü.İstatistiksel değerlendirmelerdeyse, BaP grubu seminifer tübül çaplarının, diğer gruplara karşın daha düşük olduğu saptandı. Bunun spermatogenezdeki bozulma sonucunda seminifer tübül çaplarının azalmasına bağlı olabileceği düşünüldü.Sonuç olarak, çevresel kirliliğe neden olan önemli maddelerden biri olan BaP testiste yapısal ve hücresel hasarlar ile hormonal değişimlere neden olmaktadır, bu hasar Curcumin'in antioksidan etkisiyle giderilebilir.Anahtar Kelimeler: Benzo(a)Piren, Curcumin, Testis, Histoloji The aim o f this study was to investigate the possible protective effects of Curcumin, as an antioxidant, on damage caused by BAP in testis, which is an important environmental pollution factor, using immunohistocehemical methods.This experiment used 36 Wistar Albino male rats and they were separated into six groups, each including 6 rats. Group 1: Control, Group 2: Corn oil, Group 3: DMSO, Group 4: BaP, Group 5: Curcumin, Group 6: BaP+Curcumin.After 6 weeks, testis were extracted and prepared for investigation by light microscopy. Some of the sections were stained by Hematoxylin Eosin, and the others were stained by indirect immunohistochemical methods using AT1 and Androgen Receptor (AR) primer antibodies to follow the changes in Leydig cells and the distribution of ARs, respectively. Immunoreaction of AT1 as an important marker for seminiferous epithel and particularly Leydig cells during spermatogenesis and spermiogenesis increased by BaP administration, and it was partially returned by the administration of Curcumin. In light of these findings it was thought that BaP could impair the testosterone synthesis Core involvement of AR, which has important effects on male reproductive system was usually detected adluminal cells in the BaP group. A moderate level immunoreactivity was viewed in some incorporated spermatids. It was seen that reaction against BaP came close to the controls in the group of BaP + Curcumin, indicating BaP could negatively affect the spermiogenesis.The statistical analysis indicated that the diameters of seminiferous tubules was lower in BaP group than the other groups. This finding may reveal that diameters of seminiferous tubule may be decreased as a result of deterioration in spermatogenesis. In conclusion, BaP as one of important enviromental pollution factors can cause structural and cell damage in the testis and can also creates hormonal changes. These detrimental effects of BaP can be avoided by using Curcumin as an antioxidant.Key Words: Benzoapyrene, Curcumin, Testes, Histology 156
