Amaç: PKOS, hiperandrojenizm, oligo/anovulasyon ve ultrasonografik görüntüsünde polikistik yumurtalıkların varlığı ile karakterize endokrin bir bozukluktur. Son yapılan çalışmalarda PKOS'ta oksidatif stresin etkileri üzerinde durulmaktadır. Mevcut çalışmada oksidatif strese bağlı DNA hasar göstergesi olan 8-OHdG seviyelerinin HPLC ile ölçümü ve sonuçlarının oksidan ve antioksidan sistem metabolitleri ile nasıl değiştiğinin araştırılması amaçlanmıştır.Materyal ve Metot: Bu çalışmaya PKOS tanısı almış 99 gönüllü hasta ve 47 gönüllü sağlıklı kişi dahil edildi. Alınan kan örneklerinden 8-OHdG seviyeleri HPLC ile MDA, SOD ve GPx analizleri ise spektrofotometrik olarak ölçüldü. Bulgular: 8-OHdG düzeyi ve lipit peroksidasyon ürünü olan MDA hasta grubunda kontrol grubuna kıyasla istatistiksel olarak anlamlı yüksek bulundu. Antioksidan belirteci olan SOD ve GPx ise hasta grubunda anlamlı olarak düşük bulundu. Sonuç: 8-OHdG ve MDA parametrelerinin PKOS'ta artmış olarak bulunması oksidatif stresin varlığını ve SOD ve GPx parametrelerinin azalmış olarak bulunması ise antioksidan kapasitenin yetersiz olduğunu düşündürmektedir. Aim: PCOS is an endocrine disorder, which is characterized by hyperandrogenism, oligo/anovulation, and the presence of polycystic overs in ultrasonografical evaluation. The recent studies have focused on the effect of oxidative stress on PCOS. It was aimed by the present study to investigate the levels of 8-hydroxy-2'-deoxy guanosine, a DNA damage marker, and the relationship between this marker and oxidative stress parameters.Material and Method: The volunteer patients with PCOS (n: 99) and healthy controls (n: 47) were included in the study. In blood samples taken, 8-OHdG and MDA, SOD and GPx were determined by HPLC and spectrophotometric methods, respectively.Results: 8-OHdG and MDA levels were found to be higher in patient group than the in controls, which was statistically significant. Low activities of antioxidant markers (SOD and GPx) were determined in the patients when compared with controls. Conclusion: The increased levels of 8-OHdG and MDA in patient group may be caused by oxidative stress, and the low activities of SOD and GPx in the patients by diminished antioxidant capacities in PCOS. 95
