Summary: This thesis also includes the development of rapid screening assays, exploiting cell membrane and nuclear signaling of GPCRs. We developed a method to assay intracellular calcium ions in cultured cells on 96-well plates using a fluorometer. This assay can serve as a rapid screening method for active compounds at many different GPCRs. In addition, a reporter gene assay integrating cell surface receptor and nuclear signaling to screen for novel immunosuppressant compounds was developed. This assay was applied to the analysis of FK506 in patient blood samples and the data obtained was similar to a conventional method.
