Summary: Protein higher order structure can be characterized by solution-phase amide hydrogen exchange (HDX) combined with mass spectrometry. In order to improve the spatial resolution of this approach, we investigated the utility of ECD for fragmentation of labeled peptides without causing hydrogen scrambling. Our results showed that c-type product ions can preserve structural information in some cases whereas z-type ions showed evidence of hydrogen scrambling.
