과다 소금의 섭취는 고혈압과 더불어 심혈관계 질환을 유발하는 원인이 된다. 본 연구팀의 연구결과에서는 동일한 양의 NaCl을 함유한 미네랄이 풍부한 천일염과 이를 열처리 가공한 가공염인 죽염이 미네랄이 없는 일반소금에 비해 혈압을 낮춰주는 효과가 있음을 밝힌 바 있다. 혈관 석회화는 심혈관계 질환의 발병률과 사망률에 관련한 중요한 지표로 이용되고 있으며 이에 대한 상관성이 높은 것으로 알려져 있다. 이에 본 연구에서는 만성 요독 쥐에서 다양한 종류의 소금이 혈관 석회화에 미치는 영향에 대해 조사해 보았다. 즉, 정상 쥐 (wistar rats)에 4주간 0.75% adenine과 칼슘, 인이 함유된 사료를 섭취시켜가며 혈관 석회화를 유도하고 이후 NaCl 4% 함량을 기준으로 미네랄이 없는 일반소금 (MDS), 미네랄이 풍부한 천일염 (MRS), 자죽염 (PBS), 백죽염 (WBS) 이 함유된 식이를 4주 동안 섭취시키면서 혈압 및 혈관 석회화 관련 인자들을 조사하였다. 그 결과, PBS 식이군은 소금을 섭취한 다른 식이군에 비해 혈압이 낮게 유지되었다. 각 식이군들의 대동맥 형태학적 관찰 소견으로 MDS 식이군의 대동맥 혈관 내부가 비정상적인 형태를 나타내는 것으로 관찰되었으며, PBS와 WBS를 섭취한 식이군은 MDS 식이군에 비해 대동맥에 존재하는 칼슘, 마그네슘, 인의 함량이 낮았다. 이러한 결과는 von kossa 염색법으로 시행한 조직학적 분석 결과에서도 유사하게 나타났다. 그리고 PBS와 WBS 식이군의 혈청에서 인, creatinine의 함량은 MDS 식이군에 비해 낮게 조사되었으며 혈장에서 지질과산화물인 MDA 함량도 낮았다. 또한 각 식이군의 대동맥을 대상으로 혈관석회화 유도인자인 Runx-2와 염증반응 및 세포자멸사 경로에 관여하는 NF-kB p65, caspase-3 그리고 cleaved caspase-3 의 발현량을 western blotting 방법에 의해 조사해 보았다. 결과에 의하면 PBS와 WBS식이군은 MDS식이군에 비해 Runx-2, NF-kB p65, caspase-3, cleaved caspase-3의 발현량이 낮게 나타났다.이러한 결과를 종합적으로 고찰해볼 때 고염식이는 산화스트레스와 골형성 분화 및 세포자멸사를 통해 혈관석회화를 촉진시킨다고 알려져 있지만 천일염을 대나무와 함께 열처리 가공하여 제조한 죽염인 PBS와 WBS의 식이는 일반소금(MDS)과 천일염(MRS)에 비해 혈관 석회화를 줄여 줄 수 있는 소금인 것으로 시사되었다.
High salt intake causes hypertension that augments the risk of cardiovascular disease. Recently, our lab has found that mineral-rich solar salt and its roasted salt kept blood pressure lower compared to mineral-deficient salt when the rats were fed various salts containing the same amount of NaCl. Vascular calcification is associated with cardiovascular morbidity and mortality. We investigated effects of various salts on vascular calcification in adenine-induces uremic rats. The rats fed with diets containing 4% (w/w) NaCl in various salts [mineral-deficient reagent salt (MDS), mineral-rich solar sea salt (MRS), purple bamboo salt (PBS), and white bamboo salt (WBS)] for 4 weeks after the diet containing 0.75% adenine for 4 weeks. The PBS rats showed the lowest blood and pulse pressure levels. The morphological observation of aorta showed that the MDS rats had abnormal architecture, elevated vascular lumen radius, and median thickness compared to the PBS rats. High salt diet accelerated the process of sodium and water retention in uremic rats. The aortic calcium, magnesium, and phosphate (Pi) contents in the PBS and WBS rats were lower than those in the MDS rats. These results were consistent with the results of aortic calcification visualized by von Kossa staining. The PBS and WBS rats exhibited the lowest levels of serum Pi, plasma malondialdehyde (MDA), and serum creatinine than the MDS rats. The expression levels of Runx-2, NF-κB p65, caspase-3 and cleaved caspase-3 in aorta were suppressed in the PBS and WBS rats compared with the MDS rats. These results indicate that PBS and WBS generate less vascular calcification compared to MDS and MRS, although high-salt diet accelerates vascular calcification by promoting oxidative stress, osteogenic transdifferentation, and apoptosis.
