벼 (Oryza sativa L.)의 WT Japonica type Dongjin-byeo (DJ) 개체 와 thioredoxin m RNA interference 돌연변이 개체(Ostrxm RNAi)를 이용해서 thioredoxin m이 xanthophyll 회로를 조절하는 것을 연구했다. 강한 광선 하에서 WT와 Ostrxm RNAi 식물체에서의 zeaxanthin (Zea)의 축적량은 비슷하다. 하지만 3 시간 동안 암처리 한 후에는 Ostrxm RNAi의 Zea epoxidation의 속도가 WT 개체보다 많이 느려진 것을 확인하였다. Zea이 antheraxanthin (Ant)으로 변하는 단계는 Ant이 violaxanthin (Vio)으로 변하는 단계에 비해 더 억제되었다. Reactive oxygen species (ROS)에 대한 조직 화학적 분석을 한 결과, 강한 광선 하에서 hydrogen peroxide (H2O2)와 superoxide (O2•-)가 WT 개체보다 Ostrxm RNAi의 잎에서 더 많이 축적되었다는 것을 알 수 있었다. 100, 200, 300 mM의 농도범위에서 H2O2를 처리한 결과 300 mM의 농도에서 in vitro Zea epoxidation의 속도가 느려진다는 것을 확인하였다. 강한 빛으로 조사한 시간을 연장한 것과 Zea epoxidation를 억제한 것이 정비례가 되었다. 이 것이 Ostrxm RNAi 식물체에서만 발견되고 WT에서는 발견하지 않았다. Trx m이 없는 Ostrxm RNAi 식물체에서는 antioxidant 효소를 비활성화시킨 후 ROS를 많이 축적하기 때문에 Zea epoxidation를 억제하는 기능성이 굉장히 높은 것으로 보인다.
The possible regulation of xanthophyll cycle by thioredoxin m was studied using rice (Oryza sativa) wild type (WT) and its thioredoxin m RNA interference mutant plants (Ostrxm RNAi). During illumination of high light, the rate of zeaxanthin (Zea) accumulation was similar between the two plants, but the rate of its epoxidation in darkness for 3 h in Ostrxm RNAi was much slower than that of WT. Interestingly, the epoxidation step from Zea to antheraxanthin (Ant) was more significantly depressed than that of Ant to violaxanthin (Vio). Histochemical assays of reactive oxygen species revealed that both of hydrogen peroxide (H2O2) and superoxide (O2•-) accumulated more in the leaves of Ostrxm RNAi compared with those of WT. During high-light illumination, increasing the time of HL-illumination lead to accumulated more ROS. Treatment of H2O2 up to 300 mM could reduce the rate of in vitro Zea epoxidation. Increasing the time of per-illumination lead to increase the percentage of decreasing of Zea in direct proportion. It was observed in Ostrxm RNAi, but not in WT. The possible inhibition of Zea epoxidation by the accumulated ROS was suggested to result from the blockage of activation process of antioxidant enzymes in Ostrxm RNAi plants.
