양전자방출단층촬영기(PET)는 전임상 및 임상에서 널리 사용되고 있는 핵의학 의료영상기기로, 생체에 주입한 방사성 동위원소가 양전자방출 후 소멸반응에 의해 180도 방향으로 방출된 511 keV 에너지를 갖는 두 개의 감마선을 검출함으로써 생체내부의 3차원 영상을 제공하는 진단기기이다. 광전자증배관(PMT)은 높은 이득과 안정성 때문에 PET 검출기 광센서로 널리 사용되고 있으나, 최근에는 애벌란치 광다이오드(APD)와 가이거 모드 애벌란치 광다이오드(GAPD)들과 같이 동작전압이 낮고, 잡음이 낮으며 소형화가 가능한 반도체 광센서에 연구가 활발하게 이루어지고 있다. 또한, MR 호환성 및 가격 경쟁력이 우수하여 광전자증배관을 대체할 수 있는 미래형 PET 광센서로써 주목 받고 있다. PET/CT, SPECT/CT 등과 같은 이중영상기기의 유용성이 부각되면서, 최근에는 PET과 MRI를 결합한 PET/MRI 이중영상기기에 대한 관심이 높아지고 있다. MRI는 CT와 같이 고해상도 해부학적 영상을 얻을 수 있는 영상기기로, 방사선 피폭이 없고, 연조직 대조도가 높으며 생화학적 특성을 나타내는 영상을 획득할 수 있는 장점이 있다. 특히, PET-MRI는 동시영상획득이 가능하여 뇌신경계 연구분야나 환자 움직임 보정 등이 가능한 추가적인 장점을 가지고 있다. 이와 같은 장점으로 인하여 PET/MRI 이중영상기기에 대한 연구가 지속적으로 진행되고 있다. 본 연구실에서는 세계 최초 가이거모드 애벌란치 광다이오드 배열을 사용한 MRI 호환 뇌 전용 PET을 성공적으로 개발하였다. 지멘스가 개발한 애벌란치 광다이오드 기반의 상용 MRI 호환 뇌 전용 PET과 비교하여, 우리의 뇌 전용 PET는 온도 변화에 따라서 안정적이기 때문에 보다 좋은 시간분해능을 획득할 수 있었다. 또한, 비용, 시스템 복잡성과 개발 기간이 크게 감소했다. 본 논문은 가이거모드 애벌란치 광다이오드를 사용한 MRI 호환 뇌 전용 PET 개발을 위해 사용된 디지털 신호처리(DSP)방법과 데이터 획득(DAQ) 시스템 대하여 서술한다. 본 연구에서는 PET 검출기 출력신호로부터 감마선 검출시간정보를 보다 쉽고 정확한 검출을 위해, 개선된 디지털 신호처리방법을 개발했다. 프리러닝 ADC와 FPGA을 결합한 데이터획득보드에 감마선 신호처리방법을 적용하였고, PET 검출기 성능평가를 위해 에너지분해능과 시간분해능을 획득하였다. 또한, 다수개의 데이터획득보드를 연동함으로써 사용 가능한 데이터획득시스템을 개발하였고, 뇌 전용 PET 스캐너와 결합하여 성능평가를 수행하였다. 또한, 동시정렬 및 영상재구성 프로그램을 개발하고, 이를 PET 단층영상 획득에 적용하였다. 개발한 디지털 신호처리방법으로 우수한 에너지분해능과 시간분해능을 획득하였고, 이는 선행연구방법들과 비교하여 그 성능이 우수함을 검증하였다. 개발한 데이터획득시스템은 뇌 전용 PET 스캐너와 결합하여 PET 시스템을 구성하였고, 1152채널을 가진 GAPD 출력신호를 동시 데이터 획득 및 디지털 신호처리 할 수 있음을 검증하였다. 뇌 전용 PET의 에너지분해능, 시간분해능, 공간분해능, 민감도 등을 획득함으로써 개발한 PET 시스템 및 데이터획득 시스템의 우수성을 검증하였다. 본 연구에서 개발된 디지털 신호처리방법과 데이터획득시스템은 고성능 PET/MRI 이중영상기기를 개발에 적용하는 것이 가능하고, 그 신뢰성과 유용성이 검증됨을 시사한다.
Positron emission tomography (PET) is a nuclear medicine imaging instrumentation which provides three-dimensional images of functional processes in the body (animal or human) by detecting pairs of gamma rays. Currently, photomultiplier tubes (PMT) combined with scintillators (BGO, LSO, LYSO, and so on) were widely used as PET detectors for commercial PET systems. Comparing to PMT, recently developed avalanche photodiodes (APDs) and Geiger-mode avalanche photodiodes (GAPDs) had many advantages such as compactness, low bias voltage, low noise and MRI compatibility. The price for such semiconductor photo sensors reduced rapidly, which shows big potential for them to replace PMT in the near future. Multi-modality imaging instrumentations such as PET/CT and PET/MRI provided both functional and anatomical images which can greatly improve the accuracy of disease diagnosis than using single imaging modality. PET/CT has been already widely used all around the world, however simultaneous PET/CT imaging was impossible, CT imaging had poor contrast on soft tissue part and high radiation dose in CT imaging was harmful for patient’s health. Recently, developments of hybrid PET/MRI systems became very attractive because simultaneous PET/MRI imaging was possible (reducing scanning time), MRI has much better contrast on soft tissue part and no radiation dose was needed for MRI imaging. Our lab successfully developed the world first MRI compatible brain PET using GAPD arrays. Comparing to APD-based commercial MRI compatible brain PET developed by Siemens, our brain PET was more stable on temperature variation thus better system timing resolution was obtained. Furthermore, costs, system complexity and developing duration were heavily reduced. This thesis will present the development of digital signal processing (DSP) methods and data acquisition (DAQ) system for the MRI compatible brain PET using GAPD arrays. We developed a simple and improved digital timing method for accurate detection of PET signal arrival time. By using free-running ADC and FPGA-based gamma ray signal processing method, PET system’s performance such as energy resolution, timing resolution and sensitivity can be improved. Multiple DAQ cards-based PET DAQ system was developed to simultaneously acquire and process 1152 channels pre-amplified GAPD signals from the PET scanner. DAQ software such as DAQ control program, coincidence sorting program and image reconstruction program were successfully developed. To evaluate the developed DSP methods and DAQ system for brain PET, coincidence timing resolution for PET was measured using different timing methods, and brain PET basic performance such as energy resolution, timing resolution, sensitivity and spatial resolution were measured. The DSP methods and DAQ system developed in this study were proved reliable, feasible and useful for the design of high performance hybrid PET/MRI system.
