Chitin deacetylase is a key enzyme required for fungal cell wall integrity and stress resistance of spores through conversion of nascent chitin into chitosan. Moreover, chitin deacetylase could be potentially applied into industrial production of chitosan and chitosan oligomers with controlled degree of acetylation (DA) and pattern of acetylation (PA). For the ultimate purpose of potentially applying chitin deacetyalse towards biotechnological production of chitosan and chitosan oligomers, in this study, we for the first time isolated two extracellular chitin deacetylase-producing fungi DY-52 and DY-9 from soils and they were further identified to be Mortierella sp. and Absidia corymbifera, respectively, based on the morphological properties and the nucleotide sequence of 18S rRNA gene. The optimum culture conditions of both fungi were also investigated for a maximum chitin deacetylase production. The fungus of DY-52 exhibited maximal growth in yeast peptone dextrose (YPD) liquid medium containing 2% of dextrose at pH 5.0 and 28oC with 150 rpm. In case of DY-9, surprising to us, the chitin deacetylase production could be highly enhanced by 2% of MeOH. Thereafter, the biochemical properties of chitin deacetylase from DY-52 and DY-9 in the crude conditions were investigated. With the substrate of WSCT-50, it showed a maximal activity at pH 5.5 and 60 ℃ for DY-52 chitin deacetylase while the optimum pH and temperature were 6.5 and 55 ℃ for DY-9 chitin deacetylase. For both DY-52 and DY-9 chitin deacetylase, their enzyme activity could be strongly influenced by the addition of diverse metal ions. Both of them could be significantly enhanced in the presence of Co^(2+) and Ca^(2+) while strongly inhibited by Hg^(2+) and EDTA. The results of enzymatic deacetylation of (GlcNAc)1-7 indicated that both DY-52 and DY-9 chitin deacetylases required at least two GlcNAc residues for effective catalysis and the enzyme activity increased with the increasing of degree of polymerization of chitin oligomers. In addition, among various chitin substrates tested, WSCT-50, glycol chitin and crab chitosan (DD 71-88%) could be well handled by both DY-52 and DY-9 chitin deacetylases while other crystalline chitin substrates seemed to be hard to deacetylate. In a more detailed study, chitin deacetylase was purified and characterized from the culture filtrate of the fungus Mortierella sp. DY-52. The enzyme is an acidic glycoprotein with a pI of 4.2-4.8 and contains around 70% (wt/wt) of N-linked glycans. Enzymatic deglycosylation of the purified chitin deacetylase lead to a complete loss of activity. Its apparent molecular weight was determined to be ≈67 kDa by size-exclusion chromatography and ≈52 kDa by SDS-PAGE. The enzyme has an optimum pH of 6.0 and optimum temperature of 60oC. The enzyme activity was slightly inhibited by 1-10 mM Co^(2+) while highly inhibited by 10 mM Cu^(2+). The enzyme requires at least two GlcNAc residues for catalysis and the reaction product was confirmed by LC-MS. When (GlcNAc)6 was used as the substrate, the Km and Vmax was determined to be 1.1 mM and 54.6 µmol/min, respectively.
키틴 탈아세틸화효소는 초기 키틴을 키토산으로 전환시켜주는 역할을 통해 곰팡이 세포벽의 무결성과 포자의 스트레스 대한 저항성에 관하여 핵심역활을 한다.더욱이, 키틴 탈아세틸화효소는 아세틸화의 정도와 형태의 조절에 따라 키토산과 키토산올리고머를 산업적으로 생산하는데 이용할 수 있다.키틴 탈아세틸화효소의 산업적 이용에 있어서 궁극적인 목적은 키토산과 키토산올리고머의 생명공학적 방법을 통한 생산이다.본연구에서는 토양으로부터 키틴 탈아세틸화효소 를 생산하는 곰팡이인 DY-52 와 DY-9를 분리하였고, 이 곰팡이들을 형태적 속성과 18S rRna gene 핵산 서열 분석을 통해 동정한 결과 각각 Mortierella sp. 와 Absidia corymbifera 로 밝혀졌다. 이를 통해 이 균주들에서 처음으로 밝혀진 키틴 탈아세틸화효소 을 알수있었다. 키틴 탈아세틸화효소의 대량 생산을 위한 최적 균 배양조건을 조사한결과, DY-52 는 2% dextrose 가 포함된 peptone dextrose (YPD) 에서 pH 5.0, 28 ℃,150 rpm 에서 가장 많은 키틴 탈아세틸화효소 가 생산되었다. DY-9는 2% MeOH 를 포함한 배지에서 키틴 탈아세틸화효소 생산이 높은 특성을 나타내었다.이에 따라 DY-52 와 DY-9 로부터 생산된 키틴 탈아세틸화효소 의 생화학적인 특성을 조사하였다.DY-52 에서 생산된 키틴 탈아세틸화효소는 WSCT-50을 기질로 사용하였을 때 pH5.5 와 60℃ 에서 최대활성을 보였지만, DY-9 에서 생산된 키틴 탈아세틸화효소의 경우는 pH 6.5 와 55℃ 에서 최대활성을 보였다. DY-52 와 DY-9 의 키틴 탈아세틸화효소의 금속이온에 대한 효소 활성을 조사하였는데 Co^(2+) 와 Ca^(2+) 메 의해서는 효소활성이 급격하게 증가하였지만"C Hg^(2+) 와 EDTA 에 의해서는 활성이 저해되었다. DY-52 와 DY-9 키틴 탈아세틸화효소 에 의한 키틴올리고당(GlcNAc)_(1-7 )에 대한 효소적 방법에 의한 탈아세딜화를 위해서는 적어도 2 개의 GlcNAc 잔기가 요구되며 키틴 올리고머의 중합 정도가 증가함에 따라 활성도도 증가한다.다앙한 키틴 기질을 사용하여 실험한 결과 WSCT-50, glyc이 chitin"C chitosan (DD 71-88%)에서 DY-52 와 DY-9 에서 생산된 31 틴 탈아세틸화효소에 의해 키토산을 생산할 수 있었지만 다른 기질에서는 탈아 세틸 화 활성을 보이지 않았다. 좀 더 세부적인 실험 으로써 Mortierella sp. DY-52 배앙 후 효소를 정제하고 그 특성을 조사하였다. 그 결과 이 효소는 산성의 당단백이며 pI 값은 4.2~4.8 이고 N-linked glycans 의 비율이 70% 였다. 정제된 탈아 세틸 화 효소 를 효소적방법에 의해 deglycosylation 을 살시한 결과 탈아세틸화 활성이 없어졌다.Size-exc1usion chromatography 에 의해 측정된 효소의 분자량은 대략 67 kDa 이며 SDS-PAGE 에 의해서는 52 kDa 로 측정되었다. 이 효소의 최적 pH 는 6.0, 최적 온도는 60℃ 이다.이 효소의 활성은 1-10 mM Co^(2+) 에 의해 약하게 저해되지만 10 mM Cu^(2+)에 의해서는강하게 저해된다. 이 효소는 반응물을 생성하거나 촉진시키기 위해서 적어도 2 개의 GlcNAc 잔기가 필요한 것을 LC-MS 를 통해 확인 되었다. 키틴 올리고당(GlcNAc)_(6) 을 기질로 사용하였을 때 K_(m) 과V_(max)는 각각 1.1 mM 과 54.6 umol/min 였다.
