[目的]明确宰后LND处理对藏羊肉成熟过程中线粒体通路细胞凋亡以及肌肉嫩度的影响.[方法]选用甘南欧拉藏羊后腿肉为试验材料,用100 μmol/L氯尼达明(Lonidamine,LND)(处理组)和蒸馏水(对照组)以肉液比(m/v)1∶1 注射,4℃成熟 0、12、24、72、120、168 h,对不同成熟期线粒体膜通透性转换孔(mitochondrial permeablity transition pore,MPTP)开放程度、线粒体膜电位、胞浆Cyt-c氧化还原状态、Caspase-9活性、Caspase-3活性以及肌原纤维小片化指数(MFI)、剪切力指标进行了测定.[结果]在成熟168 h时,LND处理组的吸光值、线粒体膜电位、胞浆Cyt-c还原水平及剪切力值较对照组分别低26.4%(P<0.05)、21.63%、28.4%和15.3%.Caspase-9、Caspase-3活性、MFI值较对照组分别高30.9%、25.1%、6.5%.[结论]研究结果可为宰后藏羊肉的成熟嫩化提供依据.
[Objective]To determine the effect of LND treatment on mitochondrial pathway apoptosis and muscle tenderness during postmortem aging of Tibetan mutton.[Method]The hind leg meat of Gannan Oula Tibetan sheep was selected as the test material,and 100 μmol/L lonidamine(LND)(treatment group)and distilled water(control group)were injected at a meat-to-liquid ratio(m/v)1∶1,and the degree of MPTP opening,mitochondrial membrane potential,redox state of cytoplasmic Cyt-c,caspase-3 activity,caspase-9 activity,MFI and shear force were measured after aging for 0,12,24,72,120,168 h at 4℃.[Result]After 168 hours of aging,the absorbance,mitochondrial membrane potential,cytoplasmic Cyt-c reduction level,and shear force value of the LND-treated group were 26.4%(P<0.05),21.63%,28.4%,and 15.3%lower than those of the control group,respectively.The activity of caspase-9,caspase-3,and MFI were 30.9%,25.1%,and 6.5%higher,respectively,than that of the control group.[Conclusion]The results provided a basis for postmortem aging and tendering of Tibetan mutton.
