目的 研究醒脑静注射液对脓毒症大鼠心肌酶和线粒体呼吸链复合物的影响.方法 按照体重将SD大鼠随机分为3组:正常组10只,腹腔注射与脂多糖(LPS)等量的0.9% NaCl;其余SD大鼠腹腔注射10 mg·kg-1脂多糖(LPS),建立脓毒症模型.模型组与实验组均30只,实验组经胃管注入醒脑静注射液6.4 mL·kg-1.于给药后6,24,48 h,用试剂盒法检测不同时点各组的心肌酶和呼吸链复合物的活性,包括NADH-辅酶Q还原酶(complex Ⅰ)、琥珀酸-辅酶Q还原酶(complex Ⅱ)、辅酶Q-细胞色素C还原酶(complex Ⅲ)、细胞色素C-氧化还原酶(complex Ⅳ).结果 在24 h,实验组与模型组的血清肌酸激酶分别为(1333.20±140.87),(1813.50±243.72)U·L-1;这2组的肌酸激酶同工酶分别为(748.15±119.02),(1167.92±145.17)U·L-1.实验组的肌酸激酶、肌酸激酶同工酶水平均显著低于模型组,差异均有统计学意义(P<0.01).在24h,实验组和模型组的心肌线粒体complex Ⅰ活性分别为(2.83±0.15),(1.84±0.18)u·mg-1,这2组的complex Ⅲ活性分别为(0.91±0.13),(0.68±0.16)u·mg-1,这2组的complexⅣ分别为(19.43±1.37),(15.20±1.79)u·mg-1,实验组心肌线粒体complex Ⅰ、Ⅲ、Ⅳ活性均显著高于模型组,差异均有统计学意义(P<0.05).结论 醒脑静注射液可显著降低脓毒症大鼠心肌酶水平,提高脓毒症大鼠心肌线粒体complex Ⅰ、Ⅲ及Ⅳ活性.
Objective To observe the effect of Xingnaojing injection on myocardial enzyme and mitochondrial respiratory chain complex in sepsis rats.Methods Seventy SD rats were randomly divided into 3 groups,10 for normal group,intraperitoneal injection of the same amount of normal saline as lipopolysaccharides (LPS).The other 60 SD rats were injected intraperitoneally with 10 mg· kg-1 LPS,prepared with sterile injection water and the concentration of LPS was 1 mg · mL-1 to establish sepsis model.The 30 rats were randomly selected as model group,while another 30 rats as experimental group were injected Xingnaojing.Both model group and experimental group were randomly selected 10 rats to observe in 6 h,24 h,48 h,respectively.The levels of myocardial enzyme and respiratory chain complex (complex Ⅰ),Ⅱ,Ⅲ,Ⅳ at different times in sepsis rats were observed.Results At 24 h,in experimental and model group,the level of creatine kinase (CK) were (1333.20-± 140.87),(1813.50 ± 243.72) U · L-1,the creatine kinase isoenzyme (CK-MK) in the two groups were (748.15 ± 119.02),(1167.92 ± 145.17)U · L-1,respectively.The levels of CK and CK-MK in the experimental group were significantly lower than those in model group(P <0.01).At 24 h,in experimental and model groups,the activity of myocardial mitochondrial complex Ⅰ in the two groups were (2.83 ± 0.15) and (1.84 ± 0.18) u · mg-1,complex Ⅲ in the two groups were(0.91 ±0.13) and(0.68 ±0.16) u · mg-1,complex 1W in the two groups were (19.43 ± 1.37) and(15.20 ± 1.79) u · mg-1,respectively.The levels of myocardial mitochondrial complex Ⅰ,Ⅲ and Ⅳ in the experimental group were significantly higher than those in model group (P < 0.01) Conclusion Xingnaojing injection could significantly reduce the level of myocardial enzyme and improve the levels of complex Ⅰ,Ⅲ,Ⅳ.
